^(131)I-OC125对荷人卵巢癌裸鼠肿瘤显像的实验研究

摘  要:目的探讨131碘-抗CA125单克隆抗体(131I-OC125)在荷人卵巢癌裸鼠中的分布,为131I-OC125临床应用提供依据。方法用人卵巢癌上皮性腺癌细胞株NI H:OVCAR3分别于裸鼠腋窝、腹股沟及肩胛部皮下建立卵巢癌裸鼠皮下移植瘤模型,肿瘤长至0.5~2cm时尾静脉注射131I-OC125,对照鼠注射131碘-正常小鼠免疫球蛋白(131I-NMIgG),分别于注射后24、48、60、72、96、120h用针孔准直器行裸鼠显像,观察不同时段显像结果;每次显像结束后处死裸鼠,取各组织称重,测各组织放射性计数,计算单位质量组织的摄取率(%ID/g)及肿瘤组织与非肿瘤组织比值(T/NT)。结果18只实验鼠50个肿瘤病灶显像41个,显像率为82.0%。不同部位病灶显像率差异无统计学意义,但直径0.5~1.5cm病灶显像效果最好,显像率为88.6%(31/35);大于1.5cm病灶显像率为81.8%(9/11),但T/NT值小于直径0.5~1.5cm病灶的T/NT值;直径小于0.5cm病灶显像率为25.0%(1/4)。18只对照鼠47个瘤灶无1个显像。注射131I-OC125组不同时段T/NT值略有不同,以给药后72h T/NT值最高,但所有时间段标记抗体在肿瘤部位分布均明显高于其他部位,差异有统计学意义(P<0.01);注射131I-NMIgG组不同时段T/NT值差异无统计学意义(P>0.05)。结论131I-OC125能特异地浓聚于瘤组织中,可较好地用于卵巢癌放射免疫显像。Objective To assess the distribution of ^131I-OC125 in Nude Mice Bearing Human Epithelial Ovarian Cancer,to provide the ^131I-OC125 clinical practice with the basis. Methods We implanting separately the Human Epithelial Ovarian Cancer cell pearl NIH ,OVCAR3 in the nude mice's armpit ditch and hypoderm of the scapular region, to establish the bare mouse hypodermic transplantation model of the ovarian cancer, when the tumor proliferate to 0.5- 2cm, we injected the nude mice with ^131 I-OC125 by tail intravenous,and the comparison one injects ^131 I-NMIgG, then used the pinhole collimator for developing separately the nude mice at 24,48,60,72,96,120h after injection. The developing result of the different time interval was observed; executed the nude mice, took the organization weighing, measured each organization radioactivity counting at the every end of the developing, calculated the organization's ingestion rate of unit measurement quality(%ID/g) and the ratio of the tumor organization and the non-tumor organization(T/TN). Results Forty-one ones were developed among 50 tumor of 18 experimental mice,the incidence of the developing is 82.0 %. the incidence of the developing of the different spot tumor had no distinct disparity, but the developing effect was the best in the tumor about 1cm diameter, the incidence of the developing was 88.6 % (31/35); the incidence of the developing was 81.8%(9/11) in the tumor of the greater than 1.5cm diameter,but the T/TN value was smaller than the one of the tumor about lcm diameter; The incidence of the developing was 25.0% (1/4) in the tumor of the less than 0. 5cm diamater. In the control group,nothing was developed among 47 tumor of 18 nude mice. T/TN of the group injected with ^131I-OC125 was slightly different at different time interval,T/TN was the highest at 72h after giving the medicine, but at the all time section, the tagged immune body was distributed uniformly in the tumor spot and was obviously higher than other spots,it had distinct dispari
