摘 要:目的:利用蛋白质芯片和生物信息学方法从胃癌患者血清中筛选标志蛋白质。方法:采用表面增强激光解吸电离飞行时间质谱技术(SELDI-TOF-MS)和Q10蛋白芯片对30例胃癌患者、30例胃炎患者、20例术后胃癌患者和40例健康者血清的蛋白质指纹图谱进行了检测,使用PBSⅡ-C型蛋白质芯片阅读机读取数据,获得的结果采用CIPHERGEN公司的Biomarker wizard和Biomarker Patterns System软件分析。结果:胃癌患者与健康者血清蛋白质指纹图谱相比有18个显著差异蛋白质,其中8个蛋白质在患者血清中高表达,10个蛋白质在患者血清中低表达。三个蛋白质(5907Da、5080Da、8691Da)组合构建诊断模型可鉴别胃癌与健康者。胃癌患者与胃炎患者血清蛋白质指纹图谱相比有13个显著差异蛋白质,其中7个蛋白质在胃癌患者血清中高表达,6个蛋白质在胃癌患者血清中低表达。二个蛋白质(5907Da、6436Da)组合构建诊断模型可鉴别胃癌与胃炎患者。结论:实验证明利用蛋白质组学和生物信息学方法可以从血清中筛选出胃癌相关的标志蛋白质,可见蛋白质芯片技术对于发现和筛选血清中的胃癌标志蛋白质及疗效判断是一种有效、快速的工具。Objective To screen serum proteomie spectra for diagnostic biomarkers with surface enhanced laser desorption and ionization - time of flight (SELDI - TOF) mass spectrometry technique in patients with gastric caveinoma. Methods Serum proteomic spectra were examined with SELDI-TOF mass spectrometry and Q10 protein chip in 30 patients with gastric carcinoma, 30 patients with gastritis, 20 post - gasuectomy patients (for carcinoma) and 40 controls. Results Totally 18 distinguis hed proteomic peaks from those in controls serum were detected in serum of patients with gastric carcinoma, among which three peaks (5907Da, 5980Da, 8691Da) could be used to form a proteomie pattern for diagnosis. A total of 13 distinguished proteomic peaks from those in serum of patients with gastritis were also detected in serum of patients with gastric carcinoma and of which two peaks (5907Da, 6436Da) could be used to form the diagnostic proteomie pattern. Gonclusion SELDI -TOF mass spectometry might offer a platform for detection of diagnostic proteomic pattern for gastric carcinoma with potential wide clinical applicability.
