摘 要:目的:了解杭州地区啮齿类动物所携带的流行性出血热汉坦病毒的基因型别。方法:采用逆转录-聚合酶链反应(RT-PCR)、核苷酸序列测定和细胞培养方法对免疫荧光抗原阳性的4份鼠肺标本进行汉坦病毒分析。结果:2/4份标本获得了阳性培养物,对阳性标本的M基因组片段序列分析表明均为SEO型汉坦病毒;毒株核苷酸序列比较表明:与其他SEOV型毒株同源性均达85%以上,而与其他HNTV型毒株同源性为70%;毒株序列所构建的系统发生树显示,本次分离的二株为汉坦病毒属SEOV型,与B jHD01(DQ133505)同源性为99%,属SEOV型S3亚型。结论:杭州地区流行性出血热流行毒株可能是SEOV型汉坦病毒,且以S3亚型为主。Objective:To study the genotypes of hantavirus carrying Rodent in Hangzhou. Methods:Four hantavirus strains, which were positive by antigen immunofluorescence assay, were analyzed by reverse transcription polymerase chain reaction( RT - PCR) , nucleotide sequence and cell culture. Results:Two cell lesions sequence revealed that the segment M belongs to seoul virus. The homology of segment M was above 85% between this seoul virus and other seoul virus, and was 70% between this seoul virus and Hantaan virus. Genotype of two hantavirus strains was $3 sub - type of seoul virus, whose homology with BjHD01 (DQ133505 )was 99%. Conclusion:S3 sub - type of seoul virus is probably the main Genotypes of hantavirus strains in Hangzhou.
