摘 要:目的研究低密底脂蛋白(LDL)对大鼠肝贮脂细胞(FSC)的调控作用。方法采用放射配体结合法检测肝FSC是否存在LDL受体。通过噻唑蓝(MTT)法、透明质酸(HA)放射免疫分析法及3H-脯氨酸掺入法测定LDL对FSC增殖、HA及胶原合成的影响。结果大鼠肝脏FSC上存在特异性的、高度亲和性的及可饱和性的LDL结合位点:LDL能促进FSC增殖、HA及胶原合成。结论LDL参与FSC增殖、转化和基质合成,并可能通过受体介导影响FSC功能。Objective To study the effects of low density lipoprotein (LDL) on modulation of hepatic fat-storing cells (FSC) activity in rats . Methods Radio- ligand-binding assay were used to determine the existence of LDL receptor-binding sites on FSC surface. MTT colorimetric assay, hyaluronic acid radioimmunoassay and the technique of incorporation of 3H-proline were used to observe the effects of LDL on the proliferation, HA and collagen synthesis of FSC . Results There exist the LDL receptor-binding sites on FSC surface which were specific, highly affinitive and saturable. LDL could enhance the proliferation of FSC, as well as their synthesis of HA and collagen. Conclusion LDL are involved in the proliferation, transforma- tion and the synthesis of extracellular matrix of FSC, these can modulate the activity of FSC by their receptors.
