膀胱内翻性乳头状瘤中EphA2蛋白、mRNA的表达及意义

摘　　要：目的观察膀胱内翻性乳头状瘤(IPB)组织中受体酪氨酸激酶(EphA2)蛋白、mRNA的表达,并探讨其意义。方法采用免疫组化SP法检测26例IPB(IPB组)、15例低级别膀胱移行细胞癌(BTCC组)和10例正常膀胱组织中的EphA2蛋白。采用逆转录实时定量PCR技术检测各组EphA2 mRNA的表达。结果IPB组、BTCC组EphA2蛋白的表达量(IA值)分别为13 826±3 996和13 112±3 587,与对照组的8 364±3 915相比,P均<0.05;IPB组与BTCC组相比P>0.05。以对照组为1,IPB组与BTCC组EphA2 mRNA表达分别上调(2.12±0.95)和(2.01±0.86)倍;IPB与BTCC组比较,P>0.05。结论IPB组织中EphA2过表达,其可能促进了膀胱内翻性乳头状瘤的癌性转化。Objective To investigate the expression of EphA2 protein and mRNA in inverted papilloma of the bladder （IPB） and its significance. Methods Expression levels of EphA2 protein in 26 IPB tissues, 15 low grade BTCC tissues and 10 normal bladder tissues in control groups were determined by immunohistochemistry （SP method ） and the corresponding mRNA expression were examined by real-time reverse transcriptase polymerase chain reaction （real-time RT- PCR）. Results Comparing with in normal tissues （8 364 ± 3 915） , the protein expressions of EphA2 in IPB （ 13 826 ± 3 996） and BTCC（ 13 112 ± 3 587 ） tissues both increased greatly （P 〈 0.05 ） ; The expressions of EphA2 mRNA in IPB and BTCC tissues increased respectively about （2.12 ± 0.95） and （2.01 ± 0.86） times on the average as compared with that in normal tissues ; There were no statistical differences of EphA2 protein and mRNA expressions in between IPB and BTCC groups （P 〉 0.05）. Conclusion EphA2 overexpressions facilitats IPB transforming into cancer, EphA2 could be used as a diagnosis parameter and a novel treatment target of bladder precancerous lesions.
