摘 要:采用改良的CTAB法成功提取龙柚等6种福建省早熟良种柚叶片的DNA,并对其进行RAPD分析.从初选的100个随机引物中筛选出12个扩增效果较好的引物,对提取的DNA进行PCR扩增,获得清晰可重复的位点107个,平均每条引物产生的位点为8.9个,其中多态性位点93个,占86.9%;在部分样品中检出了特异性RAPD标记24个,占23.4%;样品间遗传相似系数比较和UPGMA聚类分析表明坪山柚和文旦柚的亲缘关系最近,度尾蜜柚和四季柚分别与其它5个柚类的亲缘关系较远.Using the CTAB way to extract the DNA from the leaves of six fine precocious pummelos in Fujian such as Longyou. Genetic relationships among them, were analyzed by RAPD (randomly amplified polymorphic DNA) technique. 12 screened 10 bases primers were used in the study. The RAPD bands of 6 precocious pummelo samples of 12 primers were recorded. They produced 107 clear and repeatable RAPD bands, in which 93 were polymorphic, accounting for 86.9%, and 24 were cultivar specific RAPD markers, accounting for 23.4%. The average amplified DNA bands by each primer were 8.9. The results of genetic similarity analysis and UPGMA clustering showed that the most similar relative relationship is the Pingshanyou Pomelo and Wendanyou Pomelo, and that the duweimiyou Pomelo and the Sijiyou Pomelo are respectively less similar relative relationship than the other five pummelos.
