摘 要:目的探讨夏枯草提取物对α-淀粉酶的抑制作用。方法采用水提和水提醇沉方法得到夏枯草水提物、浸膏、粗多糖,通过Van Loon′s碘比色法检测α-淀粉酶活性,以夏枯草水提物测定该酶动力学。结果夏枯草水提物对α-淀粉酶有抑制作用,呈量效关系,对该酶抑制类型为反竞争性抑制。夏枯草浸膏对α-淀粉酶抑制作用较水提物明显。结论夏枯草水提物、浸膏对α-淀粉酶存在抑制作用,有效部位可能在浸膏部分,为深入研究夏枯草提取物抑制单糖吸收作用提供了依据。Objective It is to explore the inhibiting function of extract from Prunella vulgaris L. on α- amylase(α- AL). Methods Aqueous extract, polysaccharide and aqueous-ethanol extract was obtained from Prunella vulgaris L. by aqueous extract and ethanol extract methods. The activity of α- AL was determined through Van Loon's method, using aqueous extract from Prunella vulgaris L. to determine its enzyme kinetics. Results Aqueous extract from Prunella vulgaris L. could inhibit the activity of α- AL, and there was a dose-effect relationship between them. The inhibiting type of this amylase was uncompetitive inhibition. The effect of aqueous extract from Prunella vulgaris L. on α- AL was more obvious than that of aqueous-ethanol extract. Conclusion Aqueous extract and aqueous-ethanol extract from Prunella vulgaris L. has inhibiting effect on α- AL,and the utility part of Prunella vulgaris L. was aqueous-ethanol extract, which provides an evidence to do further study on inhibiting glucose absorption of Prunella vulgaris L.
