摘 要:目的本实验旨在研究miR-21在白血病细胞中表达情况,探讨miR-21基因与白血病发病的关系。方法提取组织中的小于200 bp的RNA,用poly(A)聚合酶在其3′末端加尾,再用5′带40 nt延伸序列的单碱基锚定Olig-dT引物进行反转录,然后用特异引物进行real-time PCR扩增,从而检测部分miRNAs在白血病细胞株K562中的表达,并与正常人骨髓表达情况相对比。结果通过real-time PCR分析,癌基因miR-21在K562细胞中的表达量约为(1.73±0.24)×105个拷贝,明显高于正常人骨髓组织中miR-21的表达(P<0.01)。结论本研究表明miR-21在K562细胞中的表达量增高,可能在白血病的发生过程中起重要作用。Objective Studing the expression of miR-21 in leukemia cells to explore the relationship between miRo21 and development of leukemia. Methods After less than 200bp miRNA was isolated and added with poly A at 3'UTR by polymerase A, the miRNA was reversely conscripted by using 40 nt specific primers for real-time PCR at 5'end. Then, the expression of miR-21 was detected in K562 cells and bone marrow cells of health people. Results The expression level of miR-21 in K562 cell was ( 1.73 ±0. 24)×10^5 copies, obviously higher than that in health people bone marrow ceils by real-time PCR analysis ( P 〈 0. 01). Conclusion The study demonstrated that the expression of miR-21 was up-regulated in leukemia cells,which may play an important role in leukemia develop- ment.
