摘  要:目的比较在血卟啉单甲醚(HMME)介导下C666-1和CNE2鼻咽癌(nasopharyngeal carcinoma,NPC)细胞对光动力疗法(pho-todynamic therapy,PDT)的杀伤效应和引起差异的可能因素。方法利用荧光光谱测量技术和激光扫描共聚焦显微技术分别检测HMME在C666-1和CNE2细胞中的潴留浓度和亚细胞分布;用细胞克隆形成方法评估C666-1和CNE2细胞的PDT疗效。结果C666-1和CNE2细胞中HMME的潴留浓度随着细胞和光敏剂的孵育时间增加而增大,但CNE2细胞对HMME的吸收明显高于C666-1细胞;随着孵育时间的增加,细胞对HMME的吸收趋于饱和。HMME主要分布在C666-1和CNE2细胞的细胞核外周。在相同实验条件下,HMME-PDT对C666-1和CNE2细胞杀伤效果存在显著差异,CNE2的细胞存活率小于C666-1细胞。结论HMME-PDT作用下C666-1细胞存活率明显高于CNE2细胞,其主要原因是CNE2细胞对HMME的吸收高于C666-1。细胞内光敏剂的潴留浓度是决定PDT疗效的一个关键要素。Objective To compare the sensitivity of hematoporphyrin monomethyl ether (HMME) mediated photodynamic therapy (PDT) for nasopharyngeal carcinoma (NPC) C666-1 and CNE2 cell lines. Methods Laser-induced fluorescence technique and confocal microscope were used to study the uptake concentration and subcellular localizatiotl of HMME in C666-1 and CNE2 ceils, respectively. The survival rate of C666-1 and CNE2 cells after HMME mediated PDT was evaluated with colony-forming assay. Results The cellular HMME content increased with drug incubation time in early stage, and then saturated after 5 h incubation.Whereas CNE2 had an overall trend of higher uptake capability compared with C666-1. The diffuse cytoplasmic distribution was the pattern of HMME intracellular localization. There were no obviously differences in the images between C666-1 and CNE2 incubated with HMME. In terms of overall PDT dose, cell killing profiles were quite distinct for two kinds of NPC cells. C666-1 cells were less responsive to drugs. Conclusions As compared to the C666-1 cells, CNE2 cells have a higher accumulated concentration after the same incubation time. As a result, the sensitivity of HMME mediated PDT for CNE2 cells is higher than that for C666-1 ceils. The accumulated concentration of HMME has a significant impact on PDT efficiency.
