摘  要:目的探讨热应激时原代培养大鼠小胶质细胞的活化及促肾上腺皮质激素释放激素受体1(CRH-R1)表达变化规律。方法分离新生大鼠的皮层小胶质细胞进行原代培养,在不同温度热应激作用下以免疫荧光染色法检测小胶质细胞的活化率及CRH-R1表达。结果热应激可引起原代培养的小胶质细胞活化率升高,42℃时可达97%以上,38.5℃、40℃、42℃作用后CRH-R1表达增加(P<0.01),其中40℃作用4h后表达增加最明显,以上作用具有时间及温度依赖性。结论本研究结果显示热应激在体外条件下能够诱导大鼠小胶质细胞活化及CRH-R1表达增加,提示小胶质细胞在热应激反应发病机制中具有重要作用。Objective To investigate the activation and the expression of corticotrophin-releasing hormone receptor 1 (CRH-R1) in primary cultured rat mieroglias treated with different temperature. Methods The primary rat microglias were cultured and purified. The morphological changes of microglias and the changes of the expression of CRH-R1 under different temperature were observed using laser scanning confocal images. Results The rate of activated microglias was increased after heat stress ( 〉 97% at 42℃). In activated microglias, the expression of CRH-R1 protein was significantly higher in 38.5℃ , 40℃, 42℃ groups (P 〈0. 01 ) and reached a peak after 4 h at 40℃ group. Heat stress exerted a temperature and time-dependent effect on the activation and the expression of CRH-R1 in primary cultured rat microglias. Conclusion The present results suggest that microglia can be activated by the heat stress and the expression of CRH-R1 protein in activated mieroglias changes markedly when the ambient temperature is raised, which suggests that microglia activation and the expression of CRH-R1 play an important role in regulating heat stress.
