摘 要:目的研究缺血再灌注对离体鼠心线粒体心磷脂(Cardioplin)及心功能的影响。方法采用Langendorff装置建立大鼠离体心脏缺血再灌注模型,将SD大鼠随机分为正常组(NOR)和缺血再灌注组(IR),每组18例。NOR组:在Langendorff灌注平衡20min后续灌100min;IR组:平衡20min续灌30min缺血40min复灌30min。观察平衡末、缺血前及再灌注末两组不同时点心功能变化及线粒体心磷脂含量的变化。结果①心功能变化:再灌注末IR组与NOR组比较,HR、LVDP、LVEDP均有统计学意义差异(P<0.01)。②线粒体心磷脂含量的变化:再灌注末IR组线粒体心磷脂含量明显低于NOR组(P<0.01)。③再灌注末与平衡末及缺血前比较,两组心脏功能及线粒体心磷脂含量差异均有统计学意义(P<0.05或0.01)。④平衡末与缺血前比较,两组心脏功能及线粒体心磷脂含量差异无统计学意义(P>0.05)。结论缺血再灌注能引起线粒体心磷脂含量下降,导致心脏功能受损。[Objective] To study the effects of ischemia reperfusion on mitochondria cardiolipin and heart function. [Methods] Langendorff apparatus were used to establish the model of myocardial ischemia reperfusion injury. SD rats were randomly divided into two groups (eighteen rats in each group): normal group (NOR) and ischemia repeffusion group (IR). Hearts isolated from SD rats were mounted on a Langendorff apparatus and started with a 20 minutes perfusion for equilibration. NOR went on perfusion for another 100 minutes after equilibration. IR underwent 40 minutes global ischemia and followed by 30 minutes reperfusion. The following parameters weres measured:The recovery of myocardial function: heart rate(HR), left ventricular end-diastolic pressure (LVEDP) and left ventrieular developed pressure (LVDP) were measured, mitochondrial lipids were extracted and cardiolipin were determined with HPLC. [Results] When IR compared with NOR, there was worse heart function at the end of reperfusion (P 〈0.01). The cardiolipin content in NOR is higher than that of IR (P 〈0.01). At end-equilibrium and before ischemia, there were no significant differences in all datas (P 〉0.05). At the end of reperfusion and end-equilibrium or before ischemia, there were significant differences in all datas (P 〈0.05 or 0.01), [Conclusion] The content of rat myocardial mitochondria cardiolipin is decreased by ischemia reperfusion and it subsequent lead to myocardial systohe function injury.
