摘 要:目的是研究软骨多糖对小鼠Ca761乳腺癌肺转移的抑制作用及其对血管生成的影响。方法是建立615近交系小鼠乳腺癌肺转移模型,设模型组和治疗组。检测软骨多糖肺转移抑制率、肺部石蜡切片HE染色观察肿瘤侵袭情况、免疫组化CD34观察瘤组织微血管密度(MVD)以及VEGF蛋白表达。结果表明,软骨多糖肺转移抑制率达44.4%;通过肺部HE染色发现治疗组转移情况明显低于模型组;免疫组化检测结果显示治疗组CD34蛋白标记的MVD、VEGF蛋白表达水平均低于模型组。可见软骨多糖能明显抑制小鼠Ca761乳腺癌的血管生成,从而抑制肿瘤转移,与软骨多糖能够下调瘤组织VEGF蛋白表达有关。The inhibitory effects of cartilage polysaccharide on angiogenesis of mice Ca761 breast cancer cells in 615 mice were investigated. 615 Mouse models of breast cancer were established. The mice were randomly divided into two groups. Mice in the model group were injected i.p with saline, while the mice in the treatment group were injected i.p. with cartilage polysaccharide. The inlu"oitory effect of cartilage polysaccharide was analyzed by measuring the pulmonary metastasis inhibition rate. Metastasis of breast cancer cells was analyzed by lung node HE staining. The MVD changes of tumor tissues were detected by CD34 staining in vivo, and the expressions of VEGF protein were determined by immunohistochemical method. Results showed that the pulmonary metastasis inlfibition rate was 44.4%. HE staining of tumor metastasis lung node showed that, compared with the model group, the pulmonary metastasis degree of treatment group was lower. The MVD and the expressions of VEGF protein of treatment group were fewer than that of model group (P〈0.01) . It was concluded that cartilage polysaccharide could inhibit the metastasis of Ca76 l breast cancer cells in the examined 615 mice. The down-regulation of VEGF expression might play a key role in the antineoplastic effect of cartilage polysaccharide.
