急性白血病患者WAVE-1蛋白测定的临床研究

摘　　要：目的探讨WASP家族Verprolin同源蛋白-1(WAVE-1)测定在急性白血病患者中的临床意义。方法运用逆转录-聚合酶链反应(RT-PCR)和Western blotting法分别测定65例急性白血病、21例非恶性血液病及50例正常对照者血清中WAVE-1含量,并进行比较。结果急性白血病患者血清WAVE-1含量明显高于正常对照组(P<0.01);非恶性血液病患者血清WAVE-1低于急性白血病患者(P<0.01),而与正常对照组比较无显著性差异(P>0.05)。化疗后完全缓解的急性白血病患者血清WAVE-1含量明显低于化疗前(P<0.01),复发者血清WAVE-1含量又复升高(P<0.01)。结论检测WAVE-1对急性白血病诊断、鉴别诊断及预后判断均有参考价值。Objective To assess the clinical significance of detection of WASP-family verprolin-homologous protein 1 （WAVE-1） protein in patients with acute leukemia（AL）. Methods The expression of WAVE-1 in 65 AL patients,21 patients with non-malignant hematologic disease（NMHD） and 50 normal controls was detected by RT-PCR and Western blotting. Results The WAVE-1 levels in AL patients were significantly higher than normal controls （P 〈 0.01 ） ;while in patients with NMHD, the WAVE-1 was lower than the AL（ P 〈 0.01 ）, but no difference as compared with the controls （ P 〉 0.05 ）. For AL patients with complete remission after chemotherapy, the WAVE-1 level was lower than before（ P 〈 0.01 ）. Conclusion The determination of serum WAVE-1 levels is of value for reference in the diagnosis ,differential diagnosis and predicting the prognosis of acute leukemla.
