摘 要:目的探讨不同温度的热疗对人肺腺癌细胞A549/CDDP耐药逆转作用及可能机制。方法采用体外细胞培养技术,培养人肺腺癌细胞系A549及其耐药株A549/CDDP。设计不同温度(37、40、41、42、43℃)热疗2h,联合2μg/mLCDDP处理A549/CDDP细胞,检测其对化疗药物CDDP的敏感性,流式细胞仪间接免疫荧光法检测CDDP、热疗处理前后细胞中P-糖蛋白(P-gp)的表达及细胞内荧光药物CDDP的聚集量。结果A549/CDDP细胞抑制率在39~42℃范围内随热疗的温度升高而升高,42℃时最高(P<0.05)。热疗合用顺铂时,A549/CDDP细胞P-gp表达下降,胞浆内CDDP荧光强度增强。结论热疗能部分逆转人肺腺癌细胞A549/CDDP的耐药效应,其机制可能与抑制P-gp表达,增加细胞内化疗药物积聚有关。Objective To explore reversal effect and possible mechanism of thermotherapy to drug resistance of human lung adenocarcinoma cell line A549/CDDP. Methods We cultured the ordinary lung adenocarcinoma cell line A549 and A549/CDDP which had the drug resist- ance in vitro. After being treated by thermotherapy for 2 hours at 37, 40, 41,42, and 43 ~C, MTT method was used to detect the sensitivity of the cell line A549/CDDP and CDDP , meanwhile flow cytometry detection was also employed to determine the expressions of P-gp and the deposition of CDDP in A549 cell line and A549 CDDP cell line. Results The inhibition rate of A549/CDDP cells was increased with the in- crease of the temperature, and the 42 ℃ + CDDP group had the highest inhibition rate (P 〈 0.05). When combined with thermotherapy re- spectively, the positive rate of P-gp expression decreased than CDDP only, and the deposition of CDDP inside the cells increased. Conclu- sion Thermotherapy can reverse the drug resistance of lung adenocarcinoma cell line A549/CDDP, and this effect may be correlated to the inhibition of P-gp expressions and the increased accumulation of the chermical drugs inside the cells.
