摘  要:目的:研究重组人血管内皮抑制素(Endostar,恩度)对淋巴管内皮细胞的作用及淋巴管生成的影响。方法:磁珠分选小儿包皮真皮淋巴管内皮细胞进行培养,并在光镜下观察和采用细胞特异性标记物检测。设立对照组和恩度实验组,应用免疫荧光法和体外三维培养法来判定恩度对淋巴管内皮细胞有无抑制作用。结果:恩度实验组淋巴管内皮细胞骨架中微丝排列极性减弱或消失,微管无明显改变。恩度实验组淋巴管内皮细胞体外形成管状分支数量及管状结构形成能力较对照组明显下降(P<0.05)。结论:重组人血管内皮抑制素(恩度)对淋巴管内皮细胞的细胞骨架和体外成管能力有明显的抑制作用。Objective:To study the effects of recombinant human endostatin(endostar) on human lymphatic endothelial cells (hLECs). Methods:hLECs were isolated from foreskin derma by immunomagnetic beads and cultured. Then, the observation of specimens by LM and the detection of the special markers by immunofluoreseence was made. The control group, endostar experimental groups were set. Methods of the immunofluorescence and tube-like formation assay were used to examine its inhibitive effect on the lympbangiogenesis. Comparing endostar group with the control groups by the method of immunofluorescence to observe the cytoskeleton of hLECs. Results: Comparing cytoskeleton of lymphatic endothelial cells with control group, the arrangement of microfilament of hLECs in endostar group was less polar or homopolar,and microtubule has little change. The number of branches of the lymphatic tubes in endostar group were less than the control group ( P 〈 0. 05 ). Conclusion : Endostar could evidently affect the cytoskeleton of lymphatic endothelial cells and inhibit their tube-formation ability.
