摘  要:[目的]探讨MDM2、p53、p14ARF和p21WAF1蛋白在胶质瘤组织中的表达及意义。[方法]应用免疫组化SP法检测胶质瘤组织中MDM2、p53、p14ARF和p21WAF1蛋白的表达情况,并分析其与胶质瘤组织学分级之间的关系。[结果]Ⅱ级和Ⅳ级胶质瘤中,MDM2阳性表达率分别为24.00%(6/25)及56.52%(13/23)(χ2=5.298,P=0.021);p53阳性表达率分别为28.00%(7/25)及60.87%(14/23)(χ2=5.259,P=0.022)。p14ARF的阳性表达率分别为76.00%(19/25)及34.78%(8/23)(χ2=8.270,P=0.004),p21WAF1的阳性表达率分别为76.00%(19/25)及39.13%(9/23)(χ2=6.700,P=0.010)。[结论]胶质瘤组织中,MDM2和p53均呈不同程度的过度表达,且随胶质瘤恶性程度的增加而表达水平增高,但p14ARFandp21WAF1蛋白随胶质瘤恶性程度的增加而表达降低。MDM2过表达、p53突变以及p14ARF、p21WAF1蛋白表达降低与胶质瘤的演进有关。[Purpose] To investigate the expressions of MDM2, p53, p14^ARF and p21^WAF1 proteins in gliomas and their biological significance. [Methods] Expressions of MDM2, p53,p14^ARF and p21^WAF1 protein were determined in glioma by immunohistochemical streptavidin-peroxidase technique. [Results] In grade Ⅱ and grade Ⅳ glioma, MDM2 positive rates were 24.00%(6/25) and 56.52%(13/ 23) (Х^2=5.298,P=0.021); p53 positive rates, 28.00% (7/25)and 60.87% (14/23) (Х^2=5.259,P= 0.022); and p14^ARF positive rates, 76.00%(19/25)amt 34.78%(8/23)(Х^2=8.270,P=0.004), and p21^WAF1 positive rates, 76.00%(19/25) and 39.13%(9/23) (Х^2=6.700,P=0.010) respectively. [Conclusion] MDM2 and p53 protein are over-expressed to a certain extent in glioma and the positive rates elevate along with aggressive extent of glioma, but positive rates of p14^ARF and p21^WAF1 decrease along with the glioma aggression. Over-expression of MDM2, mutation of p53, and low-expression of p14^ARF and p21^WAF1 might be related to progression of glioma.
