摘  要:目的探讨黄连素对炎症介导的多巴胺能神经元损伤的影响。方法于中脑原代神经元-胶质细胞培养体系中,应用脂多糖建立多巴胺能神经元损伤的炎症机制模型,黄连素(10、20μg/ml和40μg/ml)与脂多糖(1-100 ng/ml)同时处理该培养体系。免疫细胞化学法检测酪氨酸羟化酶阳性神经元的数目,液闪测定法测定多巴胺摄取力,并检测促炎症因子,包括肿瘤坏死因子α(TNF-α)、白介素-1β(IL-1β)(ELISA法)、一氧化氮(NO)(Griess反应)和超氧化物(细胞色素c还原法)的变化。结果黄连素(10、20μg/ml和40μg/ml)对脂多糖(1-100 ng/ml)诱导的中脑多巴胺能神经元损伤具显著的保护作用,且呈剂量依赖性;黄连素也显著降低促炎症因子TNF-α、IL-1β、NO和超氧化物的生成。结论黄连素抑制促炎症因子生成,保护多巴胺能神经元。Objective To evaluate the inhibitory effect of Berberine on inflammation induced dopaminergic neuro-degeneration. Methods A model of inflammation-induced dopaminergic neuro-degeneration was established by lipopolysaccharide stimulus in mesencephalic neuron glia. Both Berberine ( 10, 20 and 40 μg/ml) and lipopolysaccharides ( 1 - 100 ng/ml) were administrated into the culture system. The number of tyrosine hydroxylase positive cells was determined by immunocytochemistry. Cell bioactivity was assessed by [3^H] DA uptake analysis. Changes of inflammatory factors such as TNF-α, IL-1β (by ELISA), NO (by Griess method) and superoxide (by cytochrome C method) were examined. Results Berberine (10, 20 and 40 μg/ml ) dose-dependently protected mesencephalic dopaminergic neuron from degeneration induced by lipopolysaccharide (1 - 100 ng/ml ). Besides, Berberine inhibited significantly the production of TNF-α, IL-1β, NO and superoxide significantly. Conclusion Berberine may protect mesencephalic dopaminergic neurons by inhibiting the production of proinflammatory factors.
