摘  要:目的探讨DNA甲基转移酶1(DNMT1)和DNA甲基转移酶3b(DNMT3b)在人膀胱癌组织和人膀胱癌细胞系BIU-87中的表达及意义。方法用免疫组织化学法分别检测DNMT1和DNMT3b蛋白在24例膀胱癌组织和10例正常膀胱组织中的阳性表达率;用RT-PCR和Western blot分别检测DNMT1和DNMT3b基因在人膀胱癌细胞系BIU-87中mRNA表达水平和蛋白表达水平。结果在24例膀胱癌组织中,DNMT1阳性表达率为(75.29±2.51)%,而在10例正常膀胱组织中的阳性表达率为(12.61±1.57)%,二者之间的差异具有显著性意义(P<0.05);DNMT3b在24例膀胱癌组织中的阳性表达率为(70.91±2.86)%,在10例正常膀胱组织中的阳性表达率仅为(10.16±1.22)%,二者差异亦具有显著性意义(P<0.05);在人膀胱癌细胞系BIU-87中,通过RT-PCR和Western blot均检测到DNMT1和DNMT3b基因mRNA和蛋白的表达。结论在膀胱癌组织中,DNMT1和DNMT3b蛋白的表达较正常膀胱组织明显增加,说明DNMT1和DNMT3b的高表达可能与膀胱癌的发生发展有关;DNMT1和DNMT3b在人膀胱癌细胞系BIU-87中存在表达。Objective To study the expression of DNA methyltransferase 1 (DNMT1) and DNMT 3b (DNMT3b) in human bladder carcinoma tissues and cell line BIU-87. Methods The expression of DNMT1 and DNMT3b proteins was detected in 24 bladder carcinoma specimens and 10 normal bladder tissue specimens by immunohistochemistry. RT-PCR and Western blot were used to detect the expression of DNMT1 and DNMT3b mRNA and protein in human bladder carcinoma cell line BIU- 87. Results The positive expression rate of DNMT1 protein was (75.29±2. 51)% and (12. 61±1.57) % in 24 bladder carcinoma specimens and 10 normal bladder tissue specimens respectively (P〈0.05). The positive expression rate of DNMT3b protein was (70.91±2.86) % and (10.16±1.22)% in 24 bladder carcinoma specimens and 10 normal bladder tissue specimens respectively (P〈0.05). The mRNA and protein expression of DNMT1 and DNMT3b was both positive in human bladder carcinoma cell line BIU-87. Conclusion The expression levels of DNMT1 and DNMT3b proteins in bladder carcinomas are significantly higher than in normal bladder tissues. This result suggests that the over-expression of DNMT1 and DNMT3b may be related to the carcinogenesis and growth of bladder carcinoma. Both DNMT1 and DNMT3b genes are expressed in human bladder carcinoma cell line BIU-87.
