摘  要:目的:观察半枝莲提取的总黄酮(ESB)在体外对人舌鳞癌SAS细胞超微结构和增殖力的影响,探讨其可能的作用机制。方法:用0.1mg/mL,0.5mg/mL,2.5mg/mL和10.0mg/mLESB分别作用于SAS细胞24~72h,用四甲基偶氮唑盐(MTT)比色法检测细胞活性;透射电镜观察细胞超微结构的变化;免疫组化P-V两步法检测凋亡相关蛋白Bcl-2,Bax,Caspase-3用药前后表达变化。结果:ESB在体外对人舌鳞癌SAS细胞增殖有抑制作用,且呈时效及量效关系;透射电镜观察到典型凋亡细胞,线粒体等细胞器的形态变化与ESB的浓度和作用时间有关;用药后凋亡相关蛋白Bcl-2表达减弱,Bax,Caspase-3表达增强。结论:ESB可影响舌癌细胞的超微结构,降低细胞的增殖力从而抑制舌鳞癌SAS细胞过度生长;ESB诱导舌鳞癌细胞凋亡,可能与下调Bcl-2,上调Bax,Caspase-3等凋亡相关蛋白表达有关。Objective : To investigate the inhibition effect of total flavonoids extracted from scutellariae barbara( total fla- vonoids extracted from scutellariae barbara, ESB) on the proliferation of human tongue cell line SAS in vitro and explore the mechanism. Methods: SAS cell was treated by serial concentration of ESB(0. lmg/ml, 0.5 mg/ml, 2.5 mg/ml, 10.0 mg/ml) for 24h to 72h. The inhibition activity of the ESB against SAS was estimated by MTY assay. Morphologic changes of the cell were observed under transmission electron microscope. Expressions of apoptosis - associated proteins, such as Bcl - 2, Bax, and Caspase - 3 were analyzed by immunohistochemisty. Results : ESB could inhibit proliferation of SAS cell in vitro in a time - and dose - dependent manner. Changes of apoptotic morphology of the cells treated by ESB, which were associated with the concentration of ESB and treated time, were observed under transmission electron microscope. Bcl - 2 expression was significantly down -regulated in response to ESB treatment whereas Bax and Caspase -3 expressions were up - regulated. Conclusion: ESB can inhibit SAS cell proliferation, and increase cell apoptosis in vitro, which may relate to the down - regulated of Bcl - 2 and up - regulated of Bax and Caspase - 3.
