摘 要:目的研究一种发病机制,病理改变与临床接近且造模周期较短的大鼠房颤模型制备方法与技术。方法大鼠连续给药,经尾静脉注射乙酰胆碱-氯化钙混合液,每次给药后记录房颤持续时间。寻找能诱发稳定房颤的给药剂量和给药时间。电生理学方法观察正常和模型大鼠心房不应期的变化。结果CaCl210 mg/mL,乙酰胆碱66μg/mL模型大鼠从第4 d起房颤时间缓慢增长,7 d后趋于稳定。与正常组相比,模型大鼠心房肌ERP显著缩短(59.3 ms±5.8 ms)。结论CaCl210 mg/mL+乙酰胆碱66μg/mL混合液连续给予7 d诱导产生的大鼠房颤模型,其电生理学的病理变化与临床房颤相似,适用于化合物抗房颤活性筛选研究。Objective To establish an artial fibrillation animal model in rats. Methods To establish the artial fibrillation model,mixed Ach-CaCl2solution was injected into the caudal veins of SD rats at different concentrations for 7 days. Electrocardiogram was recorded everyday before and after the treatment. The effective refractory period (ERP) of artial muscle was determined in the last day. Results The mixed solution of Ach-CaCl2 could induce a stable artial fibrillation at the concentration of CaCl2 10 mg/mL and Ach 66 μg/mL in rats. Duration time of artial fibrillation was enhanced at the first three days and became stable during the last four days. The ERP of artial muscle was 59.3 ± 5.8 ms in model group which was shorter than that in the normal group (77.3 ± 5.3 ms). Conclusion A stable artial fibrillation model in rats can be established by caudal vein injection of Ach-CaCl2 solution. This model is mimics the mechanism of artial fibrillation in human. It is simple and convenience to be used in screening the drugs for anti-artial fibrillation in small animals.
