摘  要:目的:探讨α-catenin和黏着斑激酶(Focal adhesion kinase,FAK)在结肠癌中的表达特点及临床意义。方法:分别应用免疫组化、Western blot方法来检测34例结肠癌患者肿瘤组织及相应正常组织的α-catenin和FAK的表达。结果:α-catenin和FAK在肿瘤组织和正常组织中均有表达,主要表达于细胞膜和细胞浆。α-catenin的表达在肿瘤组织中明显低于正常组织(t=10.693,P<0.01),FAK在肿瘤组织中的表达明显高于正常组织(χ2=10.350,P=0.001),两者在肿瘤组织中的表达呈弱负相关(Pearson相关系数为-0.383,P=0.025)。α-catenin和FAK的表达在结肠癌TNM分期、有无远处转移、和有无淋巴转移方面差异有显著意义(P<0.01),与患者的年龄、性别之间未发现关系。α-catenin的表达在肿瘤的病理组织分化程度方面差异亦有显著意义,FAK在不同肿瘤病理组织分化程度之间表达没有统计学意义。结论:肿瘤组织中FAK的高表达与α-catenin的低表达,共同在结肠癌的发生发展过程中扮演着重要角色。Objective:To investigate the expression ofα -catenin and focal adhesion kinase (FAK) in colon carcinoma and to reveal their correlation with tumor clinicopathological features. Methods:The expression of α -catenin and FAK in tumor tissue and normal tissue of 34 colon caminoma patients were detected by immunohistochemistry and Western blot.Results:The expression of α-eatenin and FAK were detected both in tumor tissue and normal tissue,mostly in plasmalemma and endochylema.The expression of α-catenin was significantly lower in tumor tissue than that in normal tissue(t=10.693, P〈0.01 ), but the expression of FAK was higher in tumor tissue than that in normal tissue( Χ^2= 10.350,P=0.001 ). A weak negative correlation was found between FAK and α -catenin (Pearson correlation coefficient=-0.383 ,P=0.025 ).The expression of α-catenin and FAK were significantly correlated with TNM staging,distant metastasis and lymph node metastasis of colon carcinoma (P〈 0.01 ), but not with the sex and age of patients.The expression of α -catenin was also related to tumor differentiation status,but the expression of FAK was not statistiealy significant in different tumor differentiation status. Conclusion:The overexpression of FAK and the reduced expression of α-catenin in tumor tissue play an important role in the tumorigenesis and progression of colon carcinoma.
