摘 要:目的:研究使用超声辐照后神经母细胞瘤株SK-N-SH细胞超微结构及P-gp表达水平的改变,探讨超声波影响SK-N-SH细胞化疗敏感性的机制。方法:将SK-N-SH细胞分为实验组和对照组,实验组细胞进行超声辐照(频率:0.8MHz,声强:1W/cm2,时间:40s),对照组细胞进行超声假照。使用扫描电镜观察实验组和对照组细胞的超微结构并拍照。使用免疫组化方法检测实验组和对照组细胞P-gp的表达。结果:(1)通过观察对比实验组和对照组细胞扫描电镜照片发现:实验组细胞形态改变;细胞膜表面有大小不等的孔;细胞微绒毛脱落、减少。(2)免疫组化结果显示:对照组SK-N-SH细胞P-gp阳性表达率为56.23%±9.86%,实验组SK-N-SH细胞P-gp阳性表达率为34.86%±6.19%,P<0.05。实验组细胞P-gp表达率较对照组明显降低。结论:超声辐照可以破坏SK-N-SH细胞的细胞膜结构以及使SK-N-SH细胞P-gp的表达降低,这导致SK-N-SH细胞化疗敏感性提高。Objective: To investigate the effect of ultrasound on the ultrastructure and expression of P-gp of Neuroblastoma cell in order to discuss the mechanisms of ultrasound affecting the chemotherapeutic sensitivity of SK-N-SH cell. Methods:The SK-N-SH cells were divided into the experimental group and the control group. In the experimental group, the cell suspensions were exposed to ultrasound irradiation. In the control group, the cell suspensions were exposed to sham irradiation. The changes of ultrastructure of tumor cells were observed by scanning electron microscope and the expression of P-gp in two groups were detected. Results: ( 1 ) In the pictures took by scanning electron microscope, we found that: the configuration of tumor cell changed and there were some holes of different diameters on the cell membrane and the numbers of microvillus reduced or disappeared after ultrasound irradiation. (2) The result of immunocyto- chemisty showed the expression rate of P-gp in SK-N-SH cells in the control group was 56.23%±9.86% vs 34.86% ± 6.19% in the experimental group (P〈0.05), which shows the expression of P-gp in SK-N-SH ceils exposed to ultrasound was lower than that of the cells exposed to sham irradiation. Conclusion: Ultrasound can damage the structure of cell membrane and reduce the expression of P-gp in SK-N-SH cells, which are the reasons for ultrasound enhancing the chemotherapeutic sensitivity of SK-N-SH cell.
