喉鳞癌中ΔNp63和ΔNp73的表达及临床意义

摘　　要：目的通过研究喉鳞癌中ΔNp63和ΔNp73的表达及与患者各临床病理参数的关系,探讨其在喉鳞癌发生、发展中的作用。方法采用免疫组化二步法,检测40例喉鳞癌及相应癌旁组织中ΔNp63和ΔNp73的表达情况。结果ΔNp63和ΔNp73在喉鳞癌组织中的阳性表达率分别为85%(34/40)和82.5%(33/40)。喉癌组织中ΔNp63和ΔNp73的表达显著高于相应癌旁组织(P<0.001)。ΔNp63在喉癌组织中的表达与肿瘤分化程度有关(P<0.05);而与患者性别、临床分期、有无淋巴结转移等无关。ΔNp73在喉癌组织中的表达与患者性别、肿瘤分化程度、临床分期、有无淋巴结转移等无关(P>0.05)。喉癌组织中ΔNp63和ΔNp73的表达无显著相关性(r=0.16,P>0.05)。结论ΔNp63和ΔNp73可能作为癌基因维持肿瘤细胞干细胞样的增殖特性,在喉鳞癌的发生、发展中发挥重要作用,并可能成为喉癌成瘤机制研究中的重要靶点。Objective To investigate the expression of △Np63 and △Np73 and their association with clinical pathology characteristics in laryngeal squamous cell carcinoma（LSCC）. Methods Expression of △Np63 and △Np73 was detected by immunehistochemistry in 40 cases of primary LSCC and matched adjacent normal tissues. Results The positive rates of △Np63 and △Np73 were 85.0% and 82.5% ,respectively in laryngeal tumors. The positive rates and expression level in primary LSCC were remarkably higher than that in matched adjacent normal tissues （P 〈 0. 001 ）. The expression of △Np63 was not correlated with gender, clinical stage and metastasis in lymphoid nodes, however it was correlated with tumor differentiation （ P 〈 0.05 ）. The expression of △Np73 was not related to gender, tumor differentiation, clinical stage and metastasis in lymphoid nodes （P 〉 0.05 ）. Expression of △Np63 and △Np73 was not in a correlation in primary LSCC（r =0. 16,P 〉0.05）. Conclusion △Np63 and △Np73 are dominant isoforms and may act as oncogene in LSCC.△Np63 and △Np73 may be the key targets in research on tumorigenesis of LSCC.
