摘 要:背景与目的:4.1蛋白是细胞膜骨架的一个基本成分,参与维持红细胞形状和机械特性。近来研究表明4.1蛋白可能与多种恶性肿瘤的发生有关。本研究旨在探讨细胞骨架4.1家族蛋白成员(4.1B、4.1R、4.1N、4.1G)在非小细胞肺癌组织中的表达和意义。方法:采用EnVision和免疫组织化学法检测147例非小细胞肺癌组织中4.1B、4.1R、4.1N、4.1G蛋白的表达,用Wilcoxon秩和检验及Spearman等级相关检验分析4.1蛋白与肺癌病理特征之间的相关性。结果:与癌旁组织相比,肺鳞癌组织中4.1B、4.1R、4.1N蛋白的表达明显减少,差异均有统计学意义(P<0.01);与癌旁组织相比,肺腺癌组织中4.1B、4.1R、4.1N、4.1G蛋白的表达明显减少,差异均有统计学意义(P<0.05)。4.1B、4.1G在鳞癌组织的表达均显著低于腺癌组织(P<0.05),而4.1R、4.1N在鳞癌组织和腺癌组织的表达差异没有统计学意义(P>0.05)。鳞癌组织中,4.1G的表达与分化程度成显著正相关(rs=0.386,P<0.01);腺癌组织中4.1B、4.1N、4.1G的表达与肿瘤分化程度呈显著正相关(rs=0.276,P<0.05;rs=0.248,P<0.05;rs=0.268,P<0.05)。肺鳞癌及肺腺癌中,4.1B、4.1R、4.1N、4.1G的表达与淋巴结转移与否、肿瘤大小、年龄和性别均无相关性(P>0.05)。结论:4.1蛋白在非小细胞肺癌组织中的表达明显低于癌旁组织,4.1B、4.1N、4.1G的表达与非小细胞肺癌的分化程度有关。Background and Objective: Protein 4.1, a component of cell membrane skeleton, plays a role in maintaining the shape and mechanical stability of erythrocytes. Recent researches showed that protein 4.1 may be associated with the development of tumors. This study was to investigate the expression and significance of membrane skeleton protein 4.1 family members (4.1B, 4.1R, 4.1N and 4.1G) in human non-small cell lung cancer (NSCLC). Methods: The expression of proteins 4.1B, 4.1R, 4.1N and 4.1G in 147 specimens of NSCLC was detected by EnVision plus immunohistochemistry. The correlations of 4.1B, 4.1R, 4.1N and 4.1G expression to clinicopathologic features of NSCLC were analyzed by Wilcoxon rank sum test and Spearman rank correlation analysis. Results. The protein levels of 4.1B, 4.1R and 4.1N were significantly lower in lung squamous cell carcinoma tissues than in adjacent normal tissues (P〈0.01). The protein levels of 4.1B, 4.1R, 4.1N and 4.1G were significantly lower in lung adenocarcinoma tissues than in adjacent normal tissues (P〈0.05). The protein levels of 4.1B and 4.1G were significantly lower in lung squamous cell carcinoma tissues than in lung adenocarcinoma tissues (P〈0.05). Protein 4.1G expression in squamous cell carcinoma was positively correlated to tumor cell differentiation (&=0.386,P〈0.01). In adenocarcinoma, the expression of proteins 4.1B, 4.1N and 4.1G were positively correlated to tumor cell differentiation (rs=0.276, P〈0.05; rs=0.248,P〈0.05; rs=0.268, P 〈0.05). The expression of protein 4.1s in squamous cell carcinoma and adenocarcinoma were not related to lymph node metastasis, tumor size, patients' age and sex (P〉0.05). Conclusions; Protein 4.1s are weakly expressed in NSCLC tissues than in adjacent normal tissues. The expression of proteins 4.1B, 4.1N and 4.1G are related to tumor cell differentiation.
