摘 要:目的观察联用乳酸杆菌和丁酸梭菌对小鼠溃疡性结肠炎(UC)的疗效并探讨其治疗机制。方法建立葡聚糖硫酸钠(Dextran sulfate Sodium,DSS)诱导的小鼠急性UC模型,观察给予乳酸杆菌和丁酸梭菌治疗后,小鼠结肠粘膜的病理改变和肿瘤坏死因子(TNF-α)和单核细胞趋化蛋白-1(MCP-1)表达的变化。结果乳酸杆菌和丁酸梭菌可明显减轻小鼠结肠粘膜的损伤;可明显抑制TNF-α和MCP-1的表达,尤以两菌合用组抑制作用最强。结论乳酸杆菌和丁酸梭菌对DSS诱导的UC有治疗作用,对TNF-α表达的协同抑制作用可能是其发挥协调治疗作用的分子机制。Objective To investigate the therapeutic effect of combination of Lactobacilli strain and Clostridium butyricum strain on experimental ulcerative colitis in mice. Methods Ulcerative colitis was induced in mice by intragastric administration of dextran sulfate sodium (DSS) ; tissue biochemical indexes, the expression of tumor necorisis factor (TNF - α), and monocyte chemoattractant protein- 1 (MOP- 1) in the colonic mucosa were detected after combined treatment with Lactobacilli strai,n and Clostridium butyricum strain. Results Lactobacilli strain and Clostridium butyricum strain significantly abbreviated the damage of colonic tissue and suppressed the expression of TNF-α and MOP- 1, moreover, there were the lightest histological damage and the lowest expression of TNF - α in combined use of Lactobacilli strain and Clostridium butyricum strain Conelusions Both Lactobacilli strain and Clostridium butyricum strain show the therapeutic effect on ulcerative colitis induced by DSS. There is lower expression of TNF - α in the combination group, which may be the molecule mechanism of the coeffect on ucerative colitis in mice.
