摘 要:目的观察预防性小脑顶核电刺激对自发性高血压大鼠实验性脑梗死后神经功能缺损评分及脑组织内GAP-43mRNA表达的影响。方法自发性高血压大鼠持续性双侧顶核电刺激60min,24h后行大脑中动脉栓死术(MCAO),而对照组仅行MCAO,两组动物在脑梗死后不同时相处死,定期测定大鼠神经运动功能评分,同时取脑组织标本送荧光定量PCR。结果在MCAO后3d及7d,FN组神经运动功能缺损评分分别为2.25±0.46和1.0±0.82较MCAO组(2.33±0.82和1.43±0.79)无明显差异,而FNs组梗死后14d为0.50±0.55,较MCAO组(1.25±0.71)明显降低(P<0.05)。脑梗死后病灶边缘区12h和3d时相点GAP-43mRNA表达水平在FNs组分别为0.70±0.23及1.44±0.62和MCAO组0.57±0.25及1.18±0.42无明显差异,FNs组GAP-43mRNA在7d及14d时相点分别升至3.03±1.45和1.82±0.9,较MCAO组1.55±0.72及0.92±0.52明显升高(P<0.05)。结论预防性小脑电刺激可明显降低脑梗死后神经运动功能缺损,使梗死边缘区GAP-43mRNA的表达上调,可能与促进神经可塑性有关。Objective To study the effects of cerebella fastigial nucleus stimulation (FNs) on GAP43 mRNA expression in cerebral cortical tissue in post cerebral infarction SHR. Methods 68 of 3 month-old male spontaneous hypertensive rats ( SHRs ), weighing 200 - 250 g was employed. The middle cerebral artery occlusion (MCAO) model was duplicated. The total 60 rain persistent FNs was given, then MCAO operation was conducted 24 h later in FNs group and in MCAO group animal only experienced the MCAO operation. The neurological deficiency was recorded every week following the MCAO. The experiment animal was decapitated at certain time points and brain tissue samples were taken for quantitative RT- PCR analysis for GAP43 mRNA expression. Results 3 and 7 d after MCAO, the neurological deficiency score in FNs group (2. 25 ±0. 46 and 1.0 ±0. 82 respectively) had no significant difference with those (2.33 ±0. 82 and 1.43 ± 0. 79 ) at same time point in MCAO group. It was decreased to 0. 50 ± 0. 55 at 14 d after MCAO in FNs group , which was significantly different from 1.25 ±0. 71 in MCAO group ( P 〈 0. 05). The level of of GAP-43 mRNA expression in FNs group at 12 h and 3 d posterior of MCAO (0. 70±0. 23 and 1.44 ± 0. 62, respectively) had no significant difference with those (0. 57 ± 0. 25 and 1.18 ± 0. 42 ) in MCAO group(P 〉0. 05). At 7 d and 14 d time point, they reached the height of 3.03 ±1.45 and 1.82 ± 0. 9 respectively in FNs group, significantly higher than 1.55 ± 0. 72 and 0. 92 ± 0. 52 in MCAO group ( P 〈 0. 05 ). Conclusions FNs could significantly elevate the expression of GAP43 mRNA in post cerebral infarction brain tissue. It is possibly to have a role in post stroke behavior improvement.
