摘 要:目的:为了研究乳腺癌发生的分子遗传学机理,分离克隆模型鼠乳腺癌组织中特异表达的基因。方法:利用抑制消减杂交技术,结合反向cDNA斑点杂交试验,小鼠乳腺癌组织中分离出与正常乳腺组织差异表达的EST,应用生物信息学方法鉴定所得EST的小鼠同源基因,并分析其与乳腺癌发生的相关性。结果:获得20个在小鼠乳腺癌组织中高表达的,与已知的小鼠基因片段高度同源的基因,及2个与数据库中提交的假想蛋白基因高度同源的基因。结论:利用抑制消减杂交技术,可以高效高通量分析在乳腺癌中特异表达的基因,探讨乳腺癌发生的分子机制。Objective In order to study the molecular mechanism of breast cancer, to screen the genes differentially expressed in breast cancer. Methods Suppression subtractive hybridization (SSH ) combine reverse dot blotting was performed to isolate the differentially expressed genes between breast cancer and normal mammary tissues. A subtractive cDNA library of highly expressed genes in breast tumor tissue was established. Result About 22 differentially expressed genes were obtained. By sequencing and BLAST analysis we found 2 genes are new genes. Conclusion SSH is an effective method with high specificity to detect speeific expressed genes in breast cancer genesis and development.
