从诱导细胞凋亡机理探讨中药“抑肺饮”对肺腺癌细胞A549裸鼠移植瘤抑瘤作用的实验研究

摘　　要：目的:观察中药"抑肺饮"对肺腺癌细胞A549裸鼠移植瘤的抑制作用,探讨其细胞凋亡的作用机制。方法:将肺腺癌细胞A549接种于裸鼠腋窝皮下,形成移植瘤,实验分为阴性对照组,阳性对照组,"抑肺饮"高、中、低剂量治疗组,采用流式细胞术检测CD95(Fas)、Bcl-2,免疫组织化学法检测Caspase-3,TUNEL法检测凋亡指数。结果:"抑肺饮"可明显抑制移植瘤的增长,升高凋亡指数,显著下调Bcl-2的表达,并上调CD95(Fas)及Caspase-3的表达,与阴性对照组比较,有统计学差异(P<0.05),中剂量组与鸦胆子油对照组效果相当(P>0.05)。结论:中药"抑肺饮"对肺腺癌细胞A549裸鼠移植瘤具有抑制作用,其机制可能与其诱导细胞凋亡有关。Objective： Observing the depressant effect of Chinese formula Yifei decoction to the transplantation tumor of human lung adenocarcinoma A549 cells in nude mice, investigating the possible mechanism of Yifei decoction on lung cancer＇s cell apoptosis. Methods：We inoculate the lung adenocarcinoma A.549 cells on the nude mice to form transplantation tumor. The experimental rats were divided into five groups ： normal group , oleum fmctus bruceae group,high,middie ,low dose groups of Yifei decoction group. CD95 （Fas） ,Bcl -2 were detected by flow cytometry, Caspase -3 was detected by immunohistocbemical method, and the apoptotic index were detected by tunel method. Results： Yifei decoction can restrain the growth of the transplantation tumor obviously, and raise the apoptotic index significantly. The expression of CD95（Fas） ,Caspase-3 were negative control,the difference were significant （P 〈0.05）. The effect of the moderate dose up-regulated, and the expression of Bcl - 2 were down regulated. Comparing with the Yifei decoction group and the oleum fructus bruceae group were similar（ P 〉 O. 05 ）. Conclusion：Yifei decoction can restrain the transplantation tumor of human lung adenocarcinoma A549 cells in nude mice, and the mechanism may be related to inducing cell apoptosis.
