摘  要:目的:探讨前列腺癌微环境中癌相关成纤维细胞(carcinorna-associated fibroblasts,CAFs)对非雄激素依赖性前列腺癌细胞株Du-145生物学行为的影响。方法:原代培养前列腺癌CAFs并制备CAFs条件培养基(CAFs-CM),通过增殖实验、平板集落形成实验、侵袭实验等体外实验,观察CAFs对前列腺癌细胞生物学行为的影响。结果:由从人新鲜前列腺癌组织中分离培养CAFs制备的CAFs-CM剂量依赖性地提高了Du-145细胞的增殖能力,与SFM相比,0.5 g/L的CAFs-CM明显增强了Du-145细胞的增殖能力,两者比较差异有统计学意义[(290.6±25.2)vs(88.4±12.1),P<0.05],其细胞增殖指数Ki-67明显提高(80%vs 20%);在CAFs-CM中培养的Du-145细胞与在DMEM中培养的细胞相比,其平板形成集落的能力明显提高,差异有统计学意义[(0.42±0.09)vs(0.23±0.03),P<0.05];在CAFs-CM中培养的前列腺癌Du-145细胞的变形迁徙破坏能力比在DMEM中培养的Du-145细胞明显增强[(162.12±9.13)vs(112.34±8.45),P<0.01]。结论:前列腺癌间质微环境中的CAFs促进了前列腺癌细胞的浸润与转移,是抗前列腺癌治疗药物设计的新靶点。Objective: To investigate the impact of cancer-associated fibroblasts(CAFs) in the microenvironment of prostate cancer on biological behavior of androgen-independent prostate cancer cell line Du-145. Methods: Prostate cancer CAFs was primary cultured and CAFs conditioned medium(CAFs-FM) was produced. Through the proliferation experiment, flat colony forming experiments and invasion assay to observe the impact of CAFs on the biological behavior of prostate cancer cells. Results:CAFs isolated from fresh human prostate cancer tissue and CAFs CM dose-dependent increased the Du 145 cell's proliferation capacity. Compared with SFM, 0.5 g/L CAFs- CM significantly enhanced the Du 145 cell's proliferation capacity, both there was a significant difference (290. 6 ± 25.2)vs(88.4± 12.1) (P〈0.05), Du 145 cell proliferation index Ki-67 was eorrespongingly significantly im proved (80% vs 20%). Compared with cultured in DMEM, fiat panel capacity of colony-forming and the move ment capacity of deformation were markedly improved, there were significant differences(0.42 ± 0. 09)vs(0. 23 0.03)(P〈0.05) and(162.12±9.13)vs(112.34±8.45)(P〈0.01) respectively. Conclusions:CAFs in the prostate cancer stromal microenvironment promote prostate cancer cell invasion and metastasis. CAFs is a new target for drug design in the course of the treatment of prostate cancer.
