摘  要:目的:研究大鼠心肺复苏前后心脏和肾脏的α1肾上腺素能受体数量的变化,探讨复苏后心功能失常的发生机制。方法:采用窒息使大鼠全身循环骤停作为动物模型,用放射性配基结合分析方法检测α1肾上腺素能受体数量。将18只大鼠随机分为窒息10分钟组(窒息组)和窒息10分钟后心肺复苏组(复苏组)各9只。结果:复苏组大鼠复苏后30分钟,α1肾上腺素能受体数量比窒息组明显增加。2组心脏α1肾上腺素能受体分别为(37.85±4.95)pmol/g与(25.31±2.01)pmol/g(P<0.05),肾脏α1肾上腺素能受体分别为(32.77±3.90)pmol/g与(17.62±1.40)pmol/g(P<0.01);复苏后心脏α1肾上腺素能受体亲和力(1/Kd)与窒息组比较,差异无显著性,肾脏α1肾上腺素能受体亲和力在复苏后明显下降,1/Kd为(3.06±0.62)nmol/L与(1.57±0.32)nmol/L(P<0.05)。复苏后30分钟与窒息组比较,心脏和肾脏α1肾上腺素能受体数量明显增加,肾脏α1受体亲和力在复苏组下降。结论:α1肾上腺素能受体上调可能加剧了复苏后机体高交感肾上腺素能状态的形成,并对复苏后心脏功能产生不?Abstract Objective:To investigate changes in α_1adrenoreceptors (α_1 AR) in heart and kidney before and after cardiopulmonary resuscitation (CPR).Methods:3Hprazosin was used as a radioligand to determine changes in α_1 AR in rat heart and kidney.Eighteen Wistar rats were divided into asphyxiated (n=9) and CPR (n=9) groups randomly.Results:The α1 AR number in heart after CPR was greater than that of asphyxiated group 〔(3785±495)pmol/g vs.(2531±201)pmol/g,P<005〕,while no marked changes in α1 AR affinity were found between the 2 groups 〔Kd:(4.24±1.02)nmol/L vs.(2.60±0.46)nmol/L,P>0.05〕.Similarly,renal α1 AR number was significantly increased in CPR group compared to asphyxiated group 〔(32.77±3.90)pmol/g vs.(17.62±1.40)pmol/g,P<0.01〕,and the affinity of renal α_1 AR was markedly decreased after CPR 〔Kd:(3.06±0.62)nmol/L vs.(1.57±0.32)nmol/L,P<0.05〕.The α_1 AR number in rat heart and kidney after CPR appears to be significantly
