Effect of Flavones of Sea Buckthorn Leaves on IMP and Gene Expression in Broilers under Heat-stress Conditions

摘  要:试验选用健康1日龄AA肉鸡公雏600只,采用单因素完全随机分组试验设计,从中随机抽取240只,分4组,每组4个重复,每个重复15只,4组分别为基础日粮组(对照组)、试验组Ⅰ(基础日粮+0.025%沙棘叶黄酮)、试验组Ⅱ(基础日粮+0.05%沙棘叶黄酮)、试验组Ⅲ(基础日粮+0.1%沙棘叶黄酮)。第4周时进行热应激,温度为(36±2)℃,持续1周,然后温度恢复正常。在第3、4周末屠宰采样,利用高效液相色谱法(HPLC)检测不同组间的肌苷酸(IMP)含量,利用Real-time PCR方法检测不同组间的腺苷琥珀酸裂解酶(ADSL)基因表达。第4周试验组Ⅰ、Ⅱ、Ⅲ胸肌IMP含量分别比对照组极显著提高了33.94%、75.76%和49.09%(P<0.01);第4周试验组Ⅰ、Ⅲ肝脏ADSL mRNA表达量极显著高于对照组(P<0.01)。试验表明,基础日粮中添加沙棘叶黄酮后,可提高胸肌IMP含量及其相关基因ADSL mRNA的表达量,从而缓解热应激导致的肉质风味的下降。A total 240 one-day-age Arbor Acre broilers (male) were randomly allotted to 4 treatments with 4 replicates per treatment and 15 birds per replicate. The control group was fed a basal diet,the experimental group Ⅰ,Ⅱ,Ⅲ were fed the basal diet supplemented with 0.025%,0.05%,0.1% flavones of sea buckthorn leaves,respectively. The 4th week was in heat-stress conditions,the temperature was (36+2)℃. At the end of 3th and 4th weekends,muscle samples and liver samples were taken. HPLC was performed to measure the content of IMP;Real-time PCR was performed to determine the expression of ADSL gene. The contents of breast muscle IMP of group Ⅰ,Ⅱ,Ⅲ in the 4th week were significantly increased 33.94% ,75.76% and 49.09%(P〈0.01),respectively;the expression of ADSL mRNA in the 4th week was increased significantly in group Ⅰ,Ⅲ (P〈0.01). The content of IMP and expression of ADSL mRNA were increased by basal diet supplemented with flavones of sea buckthorn leaves,therefore,the decrease of meat favor caused by heat-stress was relieved.
