摘  要:目的观察锌、硒对镉中毒小鼠金属硫蛋白合成及脂质过氧化的影响,为镉中毒的研究及防治提供理论基础。方法36只昆明小鼠随机分成6组,每组6只:对照组,经口灌注生理盐水;CdCl2组,100μmol/kg的CdCl2,经口灌胃;Zn2SO4组,Zn2SO420μmol/kg经口灌胃;Na2SeO3组,Na2SeO310μmol/kg经口灌胃;CdCl2+Zn2SO4组,Zn2SO4和CdCl2交替经口灌胃;CdCl2+Na2SeO3组,Na2SeO3和CdCl2交替经口灌胃。109Cd-血红素饱和法测小鼠组织金属硫蛋白含量;试剂盒检测小鼠组织丙二醛(MDA)含量及超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性。结果单独镉、锌、硒组及Zn+Cd、Se+Cd组小鼠肝、心和肾脏MT含量均明显高于对照组,CdCl2组小鼠肝、心和肾脏MDA含量增高、SOD和GSH-Px活力降低。CdCl2+Zn2SO4组和CdCl2+Na2SeO3组小鼠肝、心和肾脏MT含量较单用CdCl2组进一步增高(P<0.05),MDA含量降低,SOD和GSH-Px活力升高。结论锌、硒增加镉中毒小鼠金属硫蛋白合成,减轻镉中毒小鼠脂质过氧化损伤。Objective To investigate the effects of zinc and selenium on metallothionein synthesis and lipid peroxidation in cadmium poisoning mice. Methods Thirty-six Kunming mice were randomized into six groups:control group, CdCl2 group, Zn2SO4 group, Na2 SeO3 group, CdCl2 + Zn2 SO4 group, CdC12 + Na2 SeO3 group. The 0.9% NaC1 solution, cadmium chloride ( 100 μmol/kg) , 20 μmoL/ kg Zn2 SO4 , 10 μmol/kg Na2 SeO3 were given by oral gavage in control group, CdC12 group, Zn2 SO4 group, Na2 SeO3 group, respectively. Zn2 SO4 (20 μmol/kg)and cadmium chloride( 100 μmol/kg)were given ahernatingly in CdC12 + Zn2SO4 group. Na2SeO3 (10 p, mol/kg) and cadmium chloride( 100 μmoL/kg)were given ahernatingiy in CdC12 + Na2 SeO3 group. The contents of metallothionein (MT)in liver, heart and kidney were determined. Malondialdehyde(MDA), the total superoxide dismutase(SOD) activities and glutathione peroxidase (GSH-Px) activities in liver, heart and kidney were measured by respective kits. Results MT contents in liver, heart and kidney were higher in CdCl2 group,Zn2 SO4 group, Na2 SeO3 group, CdC12 + Zn2 SO4 group, CdCl2 + Na2 SeO3 group than in control group (P 〈 0.01 ). Compared with control group, the content of MDA increased and the activities of SOD and GSH-Px decreased in liver, heart and kidney in CdC12 group. MT contents in liver,heart and kidney in CdCl2 + Zn2SO4 group and CdCl2 + Na2SeO3 group were significantly higher than in CdC12 group. Meanwhile,compared with CdCl2 group, MDA level in liver,heart and kidney reduced in CdCl2 + Zn2SO4 group and CdCl2 + Na2SeO3 group,while SOD and GSH-Px vitality increased. Conclusion Zinc and selenium could increase MT formation and degrade tissue hyperoxidative injury in cadmium poisoning mice.
