摘 要:用聚丙烯酰胺凝胶电泳分析酯酶同工酶,分离胶浓度为8.5%,浓缩胶为4%,实验材料为萌发的单粒去壳水稻种子。一条新的酯酶酶带1E被发现。1E在非洲栽培稻、普通野生稻、籼稻、粳稻中,显色深浅的频率既有区别又有联系,对稻的分类及演化的研究显示出重要意义。1E的显色可分为A型(十十)、B型(十)和C型(-)。A型显色极深,与对照品种日本晴一致,C型不显色,与对照品种IRGC102321一致,B型的显色居于A型和C型之间。对7个杂交组合及其F1进行调查,发现1E的A型对C型是不完全显性。用A型及C型的材料作亲本,对4个杂交组合F2代三种类型进行x2测验,表明A型:B型:C型符合1:14:1的设想。1E的显色由两个自由组合的基因位点所控制,显性等位基因间表现出加性效应,与以前发现的酯酶同工酶带均由单个基因位点控制完全不同。A型是两对显性等位基因的纯合体,C型是两对隐性等位基因的纯合体。B型为杂合体,这与实验中观察到B型显色居于A型和C型之间,有种种变化的事实相一致。Polyacrylamide gel electrophoresis was used for esterase isozyme analysis. A new esterase isozyme band, 1E, was found.Its staining was categorized into type A, type B and type C. Type A was as dark as the staining of control variety Nipponbore, type B was between type A and type C, type C had no staining as control variety IRGC102321. It was useful in studyof classification and evolution of rice. Through analyzing parents and F1 of seven cross combinations, incomplete dominanceof alleles controlling 1E staining was found. In F2 of four cross combinations, X^2 test of type A, type B: type C for 1: 14:1 was not significant. These three types were controlled by two independent gene loci and associated with additive effect ofdominant alleles. Type A and type C were homozygous genotypes of dominance and recessiveness, respectively, while typeB was theoretically the heterozygous genotypes. It is well known that each esterase isozyme band was controlled by a singlegene locus. Two independent gene loci controlling 1E staining was not reported before.
