摘 要:目的观察豆蔻提取物对人胃癌细胞裸鼠移植瘤的影响及可能的机制。方法建立24只裸小鼠胃癌原位移植瘤模型,随机分成4组(每组6只):对照组、5-FU组、豆蔻提取物组、联合组,各组按设计剂量给药,实验期间,每3 d测量移植瘤瘤径及体积,4周后,颈椎脱臼法处死,计算抑瘤率,用免疫组化法检测各组移植瘤瘤体中COX-2和VEGF的表达情况。结果成功建立胃癌裸鼠原位移植瘤模型,各组实验裸鼠体重下降明显低于对照组,各组抑瘤率分别为:5-FU组55.99%,豆蔻提取物组42.81%,联合用药组67.36%,联合用药组瘤体中COX-2和VEGF的表达明显下调。结论豆蔻提取物可抑制胃癌原位移植瘤的生长,豆蔻提取物联合5-FU可显著抑制胃癌原位移植瘤的生长,2组具有协同作用,其抑制人胃癌裸鼠原位移植瘤生长和转移的作用机制可能与下调COX-2和VEGF的表达有关。Objectives To observe the effect of fructus amomi rotundus extraction on mice model of orthotopic transplantation of human gastric cancer and probe possible mechanism. Methods The experiment is based on 24 nude mice model of orthotopic transplantation of human gastric cancer. They are divided randomly into four groups (six in each): control group; 5-fluorouracil group(5-FU group) ; fare group; combination group: 5-FU + Fare group and for each group it is administered by the designed dose. during the experiment, measure the diameter and volume of transplantation tumor per 3 days. after 4 weeks put to death all the mice with the way of cervical dislocation, then calculate rumor inhibition rate and detect the expression of COX-2 and VEGF in immunohistoehemisty way. Results Successfully build a stable orthotopic transplantation model of gastric cancer in nude mice. The tumor inhibition rate of 5-FU group, Fare group, Combination group is 55.99 %, 42.81% and 67.36 % respectively and the weights of other groups are obviously lower than that of control group,further more, so are the expressions of COX-2 and VEGF in Com- bination group. Conclusions Inhibition on the growth of tumor is evidenced by FAR but Synergistic effect is achieved by combination of FAR with 5-FU, which shows more remarkable inhibition on the tumor growth and metastasis of orthotopic xenograft, of BGC-823 cells in nude mice. The possible mechanism of action to inhibit growth and metastasis may relate with the expressions of COX-2 and VEGF.
