摘 要:目的研究组织蛋白酶D(Cath-D)、PCNA、nm23蛋白表达与宫颈鳞癌的分化、浸润及淋巴结转移的关系,并探讨Cath-D和PCNA、nm23蛋白表达的相互关系。方法应用免疫组织化学检测10例正常宫颈组织(NEC)、20例宫颈上皮内瘤变(CIN)、48例宫颈鳞癌组织(ICC)中Cath-D、PCNA、nm23蛋白的表达。结果在宫颈鳞癌组织中的阳性表达率分别为Cath-D 83.33%、PCNA79.19%、nm23蛋白77.08%,阳性表达率及表达强度与宫颈鳞癌的分化程度、浸润深度、淋巴结转移和预后有关,Cath-D与PCNA及nm23蛋白表达呈正相关。结论在宫颈鳞癌的分化、侵袭性生长及转移过程中,癌细胞中的Cath-D扮演了一个重要角色,而且在肿瘤细胞中Cath-D与PCNA、nm23蛋白可同时过高表达,具有相互协同作用可能,是宫颈鳞癌高度恶性和预后不良的重要生物学指标。Objective To study the association of Cath-D, PCNA, nm23 protein with differentiation, invasion and lymph node metastasis in cervical squamous cell carcinoma, and evaluate the relationship between Cath-D, PCNA and nm23 protein. Methods In 10 normal cervical (NEC), 20 cervical intraepithelial neoplasia(CIN) and 48 patients with cervical squamous cell carcinoma, the expressions of Cath-D, PCNA and nm23 protein were measured by means of immuhistochemistry using monoclonal antibodies against Cath-D, PCNA and nm23 protein. Results The positive rates of Cath-D, PCNA and nm23 protein in cervical squamous cell carcinoma were 83. 33%, 79.19% and 77.08% respectively. The expression of Cath-D, PCNA and nm23 protein in cervical squamous cell carcinoma were correlated with tumor grade, infiltration, lymph nodes metastasis and prognosis. The expression of Cath-D was positively correlated with the expression of PC- NA and nm23 protein. Conclusion The expression of Cath-D by cancer cells may play an important role in the differentiation, invasive growth and metastasis of cervical squamous cell carcinoma. Overexpressions of Cath-D, PCNA and nm23 protein were seen frequently in tumours. There might be regulated interaction among Cath-D, PCNA and nm23 protein. Cath-D, PCNA and nm23 protein expression are important in the determination of the degree of malignacy and prognosis and in the prediction of lymph node metastasis of cervical squamous cell carcinoma.
