摘 要:目的研究陷阱受体3(DcR3)在肝细胞性肝癌中的表达及对血管生成的影响。方法收集40例肝癌患者肿瘤组织,40例癌旁组织,20例正常肝组织,用RT-PCR法检测组织中DcR3 mRNA的表达,流式细胞技术检测组织细胞凋亡,免疫组织化学SP法检测肝组织中CD34的表达,得到微血管密度(MVD)值。结果27例(67.5%)肝癌患者可以检测到DcR3 mRNA,而癌旁肝组织及正常肝组织均未检测出DcR3 mRNA。肝癌组平均凋亡率高于癌旁组和正常对照组(P<0.05)。肝癌患者DcR3表达与性别和年龄无关(P>0.05);在较大肿瘤、TNM分期高、有淋巴结转移、肿瘤侵犯门静脉和包膜不完整的情况下表达增高(P<0.05)。肝癌组织中血管内皮细胞均有不同程度的黄染,肝癌组MVD明显高于癌旁组和正常对照组(PP<0.05)。27例DcR3阳性患者MVD值为(142.65±45.63),明显高于13例阴性患者MVD值(110.37±42.82)(P<0.05)。结论DcR3在肝癌组织中表达明显增高,可能促进肝癌发生与发展;DcR3可能在评估肝癌肿瘤分化、浸润深度、淋巴结转移,血管形成以及TNM分期中十分重要。[Objective] To study the expression of DcR3 and the effect on angiogenesis in HCC. [Methods] The expression of DcR3 was detected by RT-PCR in 40 patients with HCC, 40 normal tissues adjacent to tumor and 20 normal tissues. Flow cytometer was used to test cell apoptosis. The immunohistochemistry (S-P) was used to test the expression of CD34 in HCC, and recorded as microvessel density (MVD). [Results] DcR3 mRNA was detected in 27 (67.5%) of 40 HCC, while was not seen in tissues adjacent to tumor and normal control. The mean apoptosis rate of HCC was higher than those of tissues adjacent to tumor and normal tissues. There were no significant relationship between DcR3 and sex or age (P 〉0.05). But in situation of bigger tumor, TNM higher degree, lymphatic follicle metastasize, and portal vein encroached, the expression of DcR3 increased (P 〈0.05). The vascular endotheliocyte cell was stained yellow in HCC, and MVD was higher than those of tissues adjacent to tumorand normal tissues. MVD in DcR3 positive group was higher than that of DcR3 negative group (P 〈0.05). [Conclusion] DcR3 expression can be highly found in gastric carcinoma. The abnormal expression of DcR3 may promote tumorigenesis and progression. DcR3 may be important in evaluating the tumor differentiation, infiltration depth, lymphnode metastasis, angiogenesis and TNM staging of hepatic cell carcinoma.
