摘  要:目的观察CpG ODN对卡介苗免疫效应的影响,为其临床应用奠定基础。方法以CpG ODN为佐剂,与卡介苗通过皮下注射和黏膜途径免疫BALB/c小鼠,称量小鼠体重,检测迟发型变态反应及小鼠血清中抗结核菌素(PPD)IgG抗体效价和Th1型细胞因子IFNγ的含量。结果CpG可增强BALB/c小鼠的迟发型变态反应,提高血清中抗PPD IgG抗体效价和IFNγ的含量,且不会影响小鼠体重增长。皮下注射较黏膜免疫效果更好。结论CpG ODN可提高卡介苗的免疫效应,作为一种有效的佐剂,有望在新型结核疫苗的开发中发挥作用。Objective To observe the influence of CpG ODN on immune effect of BCG and lay a foundation of clinical application of CpG ODN. Methods B ALB/c mice were immunized with BCG using CpG ODN as adjuvant by subcutaneous anti mueosal routes respectively, then weighed, observed for delayed type hypersensitivity(DTH) and determined for the lgG titer against PPD as well as IFNγ content in sera. Results CpG enhanced the DTH and increased the IgG titer against PPD as well as IFNγ content in sera, while showed no influence on body weights of mice. Compared with that by mueosal route, the BCG + CpG ODN vaccinated by subcutaneous route showed good immune effect. Conclusion CpG ODN enhanced the immune effect of BCG, which might be used as an effective adjuvant for development of novel tuberculosis vaccine.
