检测猪圆环病毒2型抗体SPA胶体金免疫层析方法的建立

摘　　要：采用柠檬酸三钠还原法制备胶体金颗粒,并将其标记SPA,将金标SPA包被至玻璃纤维膜上,将猪圆环病毒2型(PCV-2)Cap蛋白、抗SPA抗体分别包被至硝酸纤维素膜的检测线与质控线上,建立了一种检测PCV-2抗体的胶体金免疫层析方法。检测阳性样品时,在检测线和质控线处均出现红色条带;检测阴性样品时,仅在质控线处出现红色条带。该方法与ELISA相比,其灵敏度为89.19%,特异性为100%,并且与PCV-1、猪瘟病毒(HCV)、猪伪狂犬病毒(PRV)、猪繁殖与呼吸综合征病毒(PRRSV)、猪细小病毒(PPV)阳性血清无交叉反应。Colloidal gold was prepared with the tri-sodium citrate reduction method. To develop gold immunochromatographic assay for detection of specific antibodies of porcine circovirus type 2, protein A labelled colloidal gold was sprayed onto a conjugate pad, meanwhile PCV-2 capid protein and anti-protein A antibodies were striped in the test line position and in the control line position respectively. The color was appeared both in the test line position and in the control line position when the sample was positive. The color was appeared only in the control line postion when the sample was negative. Compared with ELISA, the sensitivity and specificity of the assay were 89. 19% and 100% respectively. No cross-reactivity with PCV-1, HCV, PRV,PRRSV,PPV positive serum samples was observed.
