摘  要:目的检测hPer2蛋白在人结直肠癌中的表达,探讨其临床意义。方法取人结直肠癌及癌旁组织各38例,以免疫组织化学方法检测其中hPer2蛋白的表达,并分析其表达与临床及病理常用指标的关系。结果在结直肠癌及癌旁正常黏膜细胞中hPer2均有表达,阳性表达率分别为81.6%(31/38)及97.4%(37/38),两者间差异有统计学意义(P〈0.05)。以癌旁组织为参照,hPer2在癌组织中表达下调者占63.1%(24/38),表达上调者占5.3%(2/38),无明显区别者占31.6%(12/38),其差异有统计学意义(P〈0.01)。hPer2表达失调与结直肠癌患者年龄≤50岁、肿瘤组织学分级高、浸润深度深、有淋巴结转移、TNM分期晚相关(P〈0.05)。结论hPer2在人结直肠癌中表达下调。hPer2表达的失调甚至缺失可能与结直肠癌的生长、浸润、转移相关。Objective To detect the expression of a circadian gene, hPer2, in human colorectal carcinoma, and analyze its relation to clinico-pathological data. Methods The expression of hPer2 was detected in 38 paired colorectal carcinoma tissues and surrounding non-cancerous tissues by immunohistochemical staining. The relationship between hPer2 and several clinicopathological parameters was analyzed. Results hPer2 protein was positively stained in 81.6% (31/38) of colorectal carcinoma tissues and 97.4% (37/38) of surrounding non-cancerous tissues ( P 〈 0.05 ). Compared with the surrounding non-cancerous tissues ,the expression of hPer2 protein was decreased in 24 cases (63.1% ), and increased in 2 cases (5.3%), while no obvious difference was found in 12 cases (31.6% ) (P 〈 0.01 ). Low expression of hPer2 protein in cancerous cells was correlated with age ( ~〈50 years), pathological differentiation, depth of tumor invasion,lymph nodes metastasis, and TNM stage of colorectal carcinoma (P 〈 0.05 ). Conclusion The expression hPer2 protein was decreased in human colorectal carcinoma. There was a positive correlation between the down-regulation of hPer2 protein with the development, invasion, and metastasis in colorectal carcinoma patients.
