摘  要:目的:研究不同浓度半乳糖凝集素-1(Gal-1)对低氧去血清培养的星形胶质细胞表达和分泌脑源性神经营养因子(BDNF)的影响。方法:建立星形胶质细胞原代培养,设定10mg/LGal-1组、1mg/LGal-1组、0.1mg/LGal-1组和对照组分别予以10、1、0.1、0mg/LGal-1干预并进行低氧去血清培养,在低氧去血清培养1h、3h、6h、12h、1d、3d时通过RT-PCR、Western blotting、ELISA检测星形胶质细胞BDNF mRNA、蛋白的表达水平和培养上清液中细胞分泌的BDNF的浓度。结果:与对照组比较,10mg/LGal-1组低氧去血清培养3h~1d星形胶质细胞合成BDNF持续增加,6h~3d星形胶质细胞分泌BDNF明显增多,均以6h时最为明显;1mg/LGal-1组低氧去血清培养12h~1d星形胶质细胞合成BDNF明显增加,但分泌BDNF没有明显变化;0.1mg/LGal-1组对低氧去血清培养各时间点星形胶质细胞表达和分泌BDNF均无明显变化。结论:10mg/LGal-1能有效促进低氧去血清培养的星形胶质细胞表达和分泌BDNF;Gal-1可能通过此途径在脑缺血后起到神经保护作用。Objective: To investigate the effects of Galectin-1 (Gal-1) on expression and secretion of astrocytic BDNF after ischemia. Methods: Primary cultured rat astrocytes were treated with serum-free media and hypoxia to mimic ischemic condition. Gal-1 at different concentrations (10 mg/L, 1 mg/L and 0. 1 mg/L) was applied to the cultured astrocytes. Then, the temporal expression of BDNF mRNA and the level of BDNF in supernatant were studied by RT-PCR, western blot and ELISA respectively. Results: Compared with the control, 10 mg/L Gal-1 treatment increased the level of BDNF mRNA from 3 h to 1 d, and enhanced the secretion of BDNF from 6 h to 3 d. Compared with 10 mg/L Gal-1 treatment group, 1 mg/L and 0. 1 mg/L Gal 1 administration showed much less effect. One-day after the treatment, 10 mg/L and 1 mg/L Gal-1 upregulated the expression of BDNF protein in as- troeytes. Conclusion: 10 mg/L Gal-1 treatment can enhance the expression and secretion of astrocytic BDNF, which may provide a neuroprotective effect to brain after ischemia.
