摘  要:目的:研究神经干细胞(NSCs)移植对大鼠脊髓损伤(SCI)后轴突再生及后肢运动功能的影响,为NSCs移植治疗SCI提供实验依据。方法:5-溴脱氧尿嘧啶核苷(BrdU)法标记处于对数生长期的NSCs,采用电控SCI打击装置制作大鼠SCI模型。设立NSCs移植组、SCI组、假手术组各10只。NSCs移植组大鼠SCI后3d进行NSCs移植。应用免疫组织化学染色观察BrdU标记的移植细胞的存活、迁移情况以及NF200的表达,采用行为学(BBB)评分法观察大鼠后肢运动功能的恢复情况。结果:NSCs移植组在移植区域及其邻近区域可检测到BrdU阳性NSCs;NF200免疫阳性细胞平均光密度值较SCI组明显增高(P<0.05);NSCs移植组术后第28天NF200免疫阳性轴突数目较SCI组明显增多(P<0.01);BBB评分亦明显高于SCI组(均P<0.05)。结论:体外培养的胚胎大鼠NSCs在移植到SCI区域后可存活、迁移,参与了SCI处神经轴突的结构重建,可促进大鼠后肢运动功能恢复。Objective: To study neural stem cell (NSCs) transplantation to promote axonal regeneration and the function of hind limbs in rat after spinal cord injury. Methods: NSCs in logarithmic phage were labeled with BrdU. Spinal cord injury (SCI) model of Sprague Dawley rats was generated with electroeircuit control spinal cord in- juring device. Thirty SD rats were randomly divided into groups sham, SCI, and NSC. The NSCs were transplanted into the injured site three days after SCI. Then the NSCs labeled with Brdu and NF200 were detected by immunohistochemisty, Hind limb function of the rats was evaluated by an independent observer who is blind to the grouping u- sing BBB open field locomotion rating score. Results: NSCs can be detected in the spinal cord after transplantation. The mean OD of NF200 was higher than that in SCI group (P〈0.05). Twenty-eight days after transplantation, the number of NF200 immunostaining positive axons was increased remarkably than that in the SCI group (P〈0.01), and the BBB score of NSC group was significantly higher than that in the SCI group (P〈0.01). Conclusion: Trans- plantation of NSCs can survive in the injured site of spinal cord and promote the axonal reconstruction, and the function of the hind limb will be improved significantly in rats after SCI.
