摘 要:根据GeneBank上已发表的H3N8亚型禽流感的非结构蛋白基因(NS)序列,设计了一对特异性引物,成功地从A/mallard/SanJiang/167/2006(H3N8)中克隆到NS基因序列,该克隆基因全长860bp,编码NS1和NS2两种非结构蛋白。与其他H3N8亚型的NS基因进行了同源性比较,核苷酸同源性为65.5%~98.3%,推导的氨基酸序列同源性为65.4%~97.8%。NS基因遗传进化树形成两个分支,该分离株处于等位基因B组。另外,此毒株NS基因在263~277位没有发生15个碱基的缺失。A/mallard/SanJiang/167/2006(H3N8)株149位为丙氨酸,其不能感染哺乳动物。The cDNA of NS genes of subtype H3N8 Avian Influenza Viruses ( A/mallard/San Jiang/167/2006 (HBN8 ) ) were amplified by reverse transcription and polymerase chain reaction (RT-PCR). The amplified fragment of NS gene was 860 bp, encoding NS1 protein and NS2 protein. Results of comparative sequence analysis indicated that the nucleotide sequence of NS gene of this strain shared 65.5 % -98.3% homology with other subtypes H3N8 viruses, and putative amino acid sequence shared 65.4%- 97.8% homology with other subtypes H3N8 viruses. Phylogenetic tree of the NS gene had two separate groups, which belonged to NS group B. There was not a unique mutation of NS gene with 15 base pair deletion from 263 to 277 sites, indicating that it belongs to Allele A sublineage I. The 149th site of this strain was Ma, indicating that it cant infect mammals.
