摘 要:目的探讨轮状病毒感染新生SD大鼠肠道分泌白细胞介素(IL)-2、IL4及γ干扰素(INF-γ)浓度的变化及乳凝集素(laetadherin)对其的影响。方法10日龄清洁级SD大鼠45只,随机分为正常对照组(N组)、轮状病毒感染组(V组)和轮状病毒感染+laetadhefin组(VL组)三组,每组15只。N组予以人工喂养;V组在人工喂养的第1天同时给予轮状病毒喂养;VL组人工喂养的第1天给予轮状病毒喂养,出现腹泻症状后给予每天0.25mg lactadhefin喂养,直到腹泻停止。分别于给病毒前(T1)、腹泻最严重时(T2)及腹泻结束后(T3)各时间点取肠内容物上清,测定细胞因子IL-2、IL-4和IFN-γ浓度。结果三组大鼠小肠内IL-2、IL4和IFN-γ浓度在T1时间点差异元明显统计学意义;在T2时间点,V组和VL组IL-2、INF-γ浓度分别为(97.97±7.99)、(25.00±8.32)pg/ml和(70.00±8.27)、(20.53±2.10)pg/ml,均低于N组[(348.84±45.44)、(33.95±5.76)pg/ml](P均〈0.05);而在T3时间点,IL组IL-2、INF-γ浓度明显高于V组[(369.83±36.83)、(42.59±7.54)pg/ml与(293.59±45.64)、(31.35±3.63)pg/ml,P〈0.05],VL组与N组浓度差异无明显统计学意义。结论Lactadhefin在轮状病毒感染大鼠能提高IL-2和IFN-γ浓度,在抗轮状病毒感染中有一定的积极作用。Objective To explore the influence of lactadherin on gut contents level of IL-2. IL-4 and IFN-γ in newborn rats with rotavirus infection.Methods Forty-five ten-day-old SD rats were randomly divided into three groups: control group(group N), rotavirus infection group(group V), rotavirus infection and lactadherin group(group VL). All the rats were fed with formula,group V and group VL were fed with rotavirus( 1 × 10^6) on the first day,then group VL were fed 25 mg/d lactadherin when they started diarrhea, until recovery. Gut contents samples were taken before feeding(T1), with most serious diarrhea(T2),after recovery(T3),the level of IL-2,IL-4 and IFN-γ in the gut contents were detected by ELISA. Results At time of T1 gut contents level of IL-2.IL-4 and IFN-γ were no statistically significant differences. Gut contents level of IL-2 and INF-γ were higher in group N[ ( 348.84 ± 45.44) pg/ml, ( 33.95 ± 5.76) pg/ml] than those in group V[ (97.97 ± 7.22) pg/ml, (25.00 ± 8.32) pg/ml] and group VL[ (70.00 ± 8.27) pg/ml, (20.53 ± 2.10) pg/ml] at time of T2( P 〈 0.05 ). At time of T3 gut contents level of IL-2 and INF-3, were higher in group VL than those in group V[ (369.83 ± 36.83)pg/ml, (42.59 ± 7.54) pg/ml vs. (293.59 ± 45.64) pg/ml, (31.35 ± 3.63) pg/ml, P 〈 0.05],but the differences between group VL and group N were not statistically significant. Conclusions Lactadherin could boost gut contents level of IL-2 and INF-γ in rats with mtavirus inretctiom,it plays a certain positive role on antimtavirus infection.
