摘 要:目的探讨增强型体外反搏抗动脉粥样硬化及其内皮机制。方法12只高胆固醇血症猪随机分为高脂组和反搏组。反搏36小时后,取冠状动脉和腹主动脉进行病理形态学及超微结构观察,收集胸主动脉内皮细胞进行蛋白组学检测。结果主动脉大体苏丹Ⅲ染色及冠状动脉HE染色示高脂组斑块形成较反搏组多,反搏组斑块/内膜面积比显著少于高脂组(1.27%±0.35%比7.83%±2.39%,P<0.05)。扫描电镜示高脂对照组内皮细胞排列紊乱,大量破坏脱落,有较多血小板黏附;反搏组内皮细胞较完整,沿血流方向梭形排列,血小板黏附较少。透射电镜示高脂对照组内皮细胞变性凋亡脱落显著,内皮下大量泡沫细胞积聚,平滑肌细胞呈合成型改变,大量增生并向内膜移行;而反搏组上述改变明显减轻。反搏组内皮细胞检测到8种高表达的蛋白质。结论增强型体外反搏通过修复高胆固醇引起的内皮细胞损伤发挥抗动脉粥样硬化作用。Aim To explore the effect of enhanced external counterpulsation (EECP) against atherosclerosis and its endothelial mechanisms. Methods Twelve hypcrcholesterolcmic pigs were randomly divided into the hypedipidemia and EECP groups. After EECP,amoant to 36 hours, the coronary arteries and abdominal aortas were sampled for histopathologic and uhrastructural examination. The endothelial cells ( EC ) were collected from thoracic aorta for proteomic study. Results Compared with the hypercholesterolemic animal receiving EECP, much more plaques were found in the hypercholestcrolemic animal without EECP. The plaque/intimal area ratio of the aorta decreased significantly in animals receiving EECP ( 1.27% ± 0.35% ) versus (7.83% ± 2.39% ) in those without EECP, P 〈 0.05. Lipid deposition, endothelial damage and proliferation of smooth muscle cells were less severe in animals receiving EECP than those without EE- CP. Moreover, eight kinds of proteins were higher expressed in ECs from animals receiving EECP than from those without EECP. Conclusion EECP could prevent atherosclerotic lesion progress by repairing damaged endothelia caused by hypercholesterolemia.
