摘 要:目的:探讨益气活血中药(芎芪合剂)对局灶性脑缺血-再灌注损伤的保护作用及机制。方法:30只SD大鼠随机分为3组:假手术组、缺血-再灌注损伤模型组、芎芪合剂治疗组。建立大鼠大脑中动脉脑缺血-再灌注损伤模型,然后对各组大鼠缺血-再灌注后脑组织中的氨基酸含量进行测定。结果:对于大鼠缺血-再灌注后脑组织中兴奋性氨基酸谷氨酸(Glu)、天门冬氨酸(Asp)的含量,芎芪合剂治疗组与模型组比较,差异有统计学意义;芎芪合剂治疗组与假手术组比较,差异无统计学意义,提示芎芪合剂能够抑制大鼠缺血-再灌注后脑组织中兴奋性氨基酸Glu、Asp含量的增高,并使之趋向于正常;对于大鼠缺血-再灌注后脑组织中抑制性氨基酸谷氨酰胺(Gln)、γ-氨基丁酸(GABA)的含量,芎芪合剂治疗组与模型组、假手术组比较,差异均有统计学意义,提示芎芪合剂同时还能协同增高大鼠缺血-再灌注后脑组织中抑制性氨基酸Gln、GABA的含量。结论:芎芪合剂能够通过调整兴奋性氨基酸和抑制性氨基酸的含量,达到减轻大鼠脑缺血-再灌注后脑组织损伤的作用。Objective: To discuss the protective effect and mechanism of Yiqihuoxue Chinese Herbal Medicine (Xiongqi Composition) on amino acid contents in brain tissue of rats after cerebral ischemia-reperfusion. Methods: 30 rats were randomly divided into three groups: sham-operated group, model group and Xiongqi Composition treatment group. The ischemia-reperfusion injury model was established, then the amino acid contents in rat brain tissue of ischemia-reperfusion were determined. Results: Xiongqi Composition treatment group showed significant difference in the contents of excitatory amino acids Glu and Asp in the brain tissues after ischemia-reperfusion compared with the model group, but it showed no significant difference compared with the sham-operated group, suggesting Xiongqi Composition could inhibit the increase of excitatory amino acids Glu and Asp contents of ischemia side brain tissue induced by ischemia-reperfusion in rats, and could cause them tend to normal. Xiongqi Composition treatment group showed significant difference in inhibitory amino acides Gin and GABA in brain tissues after ischemia-reperfusion compared with model group and sham-operated group, suggesting Xiongqi Composition could also increase the contents of inhibitory amino acids Gln and GABA in brain tissues after ischemia-reperfusion. Conclusion: Xiongqi Composition can reduce cerebral ischemia-reperfusion injury by adjusting the contents of excitatory amino acids and inhibitory amino acids.
