摘 要:目的本研究旨在建立了1种检测血浆和尿液中D-乳酸盐的酶学方法并对该法进行了方法学评价。方法用改良的紫外酶促法对血浆及尿中D-乳酸盐检测分析。结果在浓度为0-500μm血浆和尿标准曲线的线性非常好(相关系数,R=0.99)。血浆和尿液批内变异系数(CV)分别为为3.0%和5.8%;血浆和尿液的批间变异系数分别为7.1%和6.8%;血浆和尿液的平均回收率分别为(103.48.2%)和(100.1±7.2)%;结论该方法具有可接受的精度和回收率,准确快速,不需要大型仪器,可适用常规检测。Objective To establish an enzymatic assay for determination of D-lactate in plasma and urine. Methods D-lactate in plasma and and in urine was assayed enzymatically with a improved UV-spectrophotometer. Results Good linearity was observed at the concentration of 0-500μM plasma and urine standard curve (correlation coefficient,R=0.99). The within-run coefficients of variations(CV)were 3.0% for plasma and 5.8% for urine. The between run CVs were 7.1% for plasma and 6.8% for urine. The mean recovery with standard deviation(S.D.) was 103.4±8.2% for plasma and 100.1%±7.2% for urine. Conclusions The method proved reliable, accurate and fast with acceptable precision and recovery.
