不同重组人造血细胞生长因子在髓系白血病骨髓细胞染色体检测中的应用

摘　　要：目的:探讨不同造血细胞生长因子对髓系白血病细胞染色体形成的能力、可能机制及其应用价值.方法:用含不同重组造血细胞生长因子(包括白细胞介素3,粒-巨噬细胞集落刺激因子和粒细胞集落刺激因子)的培养液体外分别培养22例髓系白血病的骨髓细胞24h,G显带染色观察其染色体形成情况.结果:7例急性髓系白血病(AML)中,IL-3对4例有刺激作用,GM-CSF对3例刺激作用,G-CSF对2例有刺激作用;异常核型检出率为57.1%(4/7).15例慢性髓系白血病(CML)中,IL-3对4例有刺激作用,GM-CSF对9例有刺激作用,G-CSF对8例有刺激作用;异常核型检出率为100%.结论:造血生长因子对不同患者白血病细胞染色体形成能力的影响差异很大,IL-3对AML作用较大,GM-CSF和G-CSF对CML作用较大.多数患者的骨髓细胞染色体形成不同程度地需要一种或几种造血生长因子的刺激.AIM： To explore the potential roles of different recombinant human hematopoietic growth factors in chromosomal formation in myeloid leukemia and their applicative values. METHODS： Bone borrow simples of 22 patients with myeloid leukemia were cultured respectively with interleukin-3 （IL-3）, granulocyte-maerophage colony-stimulating factor （ GM-CSF ） and granulocyte colony-stimulating factor （GM-CSF） for 24 h and chromosomal formations were observed by G-binding staining. RESULTS： In the 7 patients with acute myeloid leukemia （AML）, IL-3 stimulated chromosomal formation was found in 4 patients, that of GM-CSF stimulation in 3 patients and that of G-CSF-stimulation in 2 patients, and the ratio of abnormal karyotype was 57. 1% （4/7）. In the 15 patients with chronic myeloid leukemia（CML）, the stimulatory function was found in 4 patients by IL-3, in 9 patients by GM-CSF and in 8 patients by G-CSF, and the ratio of abnormal karyotype was 100%. CONCLUSION： The effect of different hematopoietic growth factors varies significantly on the chromosomal formation in myeloid leukemia. IL-3 has stronger effects on AML while GM-CSF or G-CSF has stronger effects on CML. But for most patients, the chromosomal formation should be stimulated by one or more hematopoietic growth factors.
