摘  要:目的通过观察消痰散结方对裸鼠人胃癌MKN-45原位移植模型端粒酶活性表达的影响,探讨其抑制胃癌细胞增殖的作用机制。方法采用OB胶粘贴法建立裸鼠人胃癌MKN-45原位移植瘤模型,随机分成模型组、消痰散结方低、中、高剂量组、5-氟脲嘧啶(5-Fu),每组8只,运用PCR-ELISA方法检测端粒酶活性的表达。结果①消痰散结方低、中、高剂量组的平均瘤重与模型组相比,差异有统计学意义(P<0.01);②消痰散结方中剂量组和5-Fu组端粒酶活性与模型组组比较明显降低,差异有统计学意义,P<0.05,消痰散结高剂量组端粒酶活性与模型组组比较差异有统计学意义(P<0.01)。结论消痰散结方抑制胃癌细胞生长,部分可能与其抑制端粒酶活性有关。Objective To discover the mechanism of Xiaotan Sanjie Fang (XTF), a traditional Chinese medicinal empirical Prescription for treating gastric cancer by observing its influence on the expression of human gastric cancer MKN-45 telomerase activity in a nude mice in situ tumor transplantation model. Methods Using OB glue paste method,the nude mice human gastric cancer MKN-45in situ transplantation tumor model was established. Animals were randomly divided into 5 groups (8 mice/every group) : the blank model group, low-dose XTF group, median-dose XTF group,high-dose XTF group, and 5-Fu group By PCR-ELISA the expression of telomerase activity was detected. Results ① There was a significant difference in mean tumor weight in the low, median and high dose XTF groups compared with the model group (P 〈 0.01 ) ② Telomerase activity in mid dose group and 5-Fu group was lower than that of normal NS group ( P 〈0. 05 ), Telomerase activity high dose group was lower than normal NS group ( P 〈 0. 01 ). Conclusion Xiaotan Sanjie Fang can inhibit the growth of gastric cancer cells, which may be related partly to the inhibition of telomerase activity.
