健脾理气方药诱导小鼠Hca-F肝癌细胞凋亡的形态学研究

摘　　要：目的观察健脾理气方药对小鼠Hca-F肝癌细胞凋亡的影响。方法建立小鼠脾虚肝郁的动物模型,再选用小鼠Hca-F腹水型肝癌细胞株,制作肿瘤动物模型,随机将其分为肿瘤对照组、中药组、西药组、中西药结合组进行光、电镜形态学观察。结果各实验组与肿瘤对照组比较,肝癌细胞体积较小,异型性不明显,病理性核分裂较少,间质血管较少。电镜下可见到细胞形态、胞质、胞核、染色质的变化,以及细胞凋亡的特异性标志—凋亡小体形成。结论健脾理气方药具有抗肝癌作用,其作用机理是通过诱导肿瘤细胞凋亡来实现的。Objective To observe the effect of Jianpiliqi herb on apoptosis of Hca - F hepatoma cells in mice. Methods Pixuganyu mouse model was successfully established, and ascites inducible hepatoearcinoma cell line Hca - F was chosen for tumor animal model. Animals were randomly divided into blank group ,western medicine group ,Jianpiliqi herb group and integrated traditional Chinese and western medicine group. And then morphologic changes were observed by light microscope （LM） and electron microscope （EM）. Results Compared with the control group, the tumor ball and the cell volume were smaller and quiet atypia. Fewer pathological karyokinesis and interstitial blood vessels were seen in experimental groups. Through the observation of EM, apoptotie body could also be found. Conclusion Jianpiliqi herb can inhibit hepatoearcinoma by means of inducing apoptosis of tumor cell.
