摘  要:目的探讨正常造血组织在全身低剂量照射后的放射生物学效应。方法将96只雌性昆明小鼠随机分为8组,暴露于0~800mGyγ射线下。分别于照射后24、48、72h处死,制备单个核细胞悬液。加γH2AX抗体染色后应用流式细胞仪检测其荧光强度。结果单染及双染法结果均显示γH2AX表达呈J型曲线,所有时间剂量组中,100mGyγ射线在48h时诱发的DSB数量最少。结论γH2AX的表达水平可以反映DNA双链断裂及修复情况。造血组织低剂量照射后可以产生兴奋效应。Objective Phosphorylation of histone H2AX(γH2AX) is a new DNA damage repair - related protein occurs rapidly in response to the presence of DNA double strand breaks. We examined the possibility that the expression of phosphorylated H2AX level of mononuclear bone marrow cell after low dose irradiation could provide information on normal hemopoietic tissue radiobiological effect to irradiation. Methods 96 Female Kunming mice were divided into eight groups, according to their exposure to 0 -800 mGy 60Co -ray irradiation. Mice were sacrificed at 24 h,48 h and72 h after LDR, to collect mononuclear bone marrow cell. γH2AX intensity were detected using Flow cytometry of γH2AX antibody - stained single cells. Results a J-shaped curve is found from the level of γH2AX expression by both of single and double staining methods. 100m Gy γ-ray induced a minimum stimulation for DSBs at 48 h after irradiation in all the groups. Conclusion γH2AX, as an indicator of DSB, has the potential to provide useful information on normal bone marrow cells response to ionizing radiation. 2. The results suggest that LDR induces hematopoietic hormesis.
